Title: | Rewriting the history: the novel expression pattern of the RGMa protein in the satellite cell |

Abstract: | Repulsive guidance molecules (RGM) are a family of multifunctional proteins made up of RGMa, RGMb, HFE2 and RGMd. Recent studies have shown that the RGMa protein is linked to the development of skeletal muscle tissue, controlling the size and mechanism of fusion of myoblasts into multinucleated myotubules. The overexpression of RGMa in the cell causes the appearance of hypertrophic cells, and its absence causes the appearance of atrophic cells. The expected pattern of expression of this molecule in the skeletal muscle cells is in the plasma membrane, but we found it in the nucleus of the muscle stem cells – the satellite cells. In this work, it was hypothesized that the presence of the RGMa protein in the nucleus of the satellite cells is driven by the intracellular domain of its canonical receptor, neogenin. Satellite cells from C57 Black extensor digitorum longus were isolated from adult mice. Afterwards, the endogenous neogenin expression was knocked down using siRNA transfection. RGMa and neogenin expressions were determined by immunofluorescence microscopy. Primary results confirmed that RGMa protein was located in the nuclei of the satellite cells, while neogenin could be found in the perinuclear region. Confocal microscopy showed that fluorescent peaks are in the middle of the z-stack concluding that RGMa is expressed in the satellite cell nuclei. Next, western blot revealed that anchored RGMa37, is possibly the isotype present in the cell nuclei and N-RGMa is in the cytoplasm of the satellite cell. Also, a new 50kDa band may be expressed alongside RGMa37. By using siRNA-Neo1, neogenin expression was silenced and RGMa could no longer be found cell nuclei, suggesting that neogenin is the carrier for RGMa novel expression in the nuclei of the satellite cells. The presence of RGMa-Neogenin in the nucleus of skeletal muscle cells will implicate in possible new biological functions for this signaling pathway, that it has not described thus far. |
Abstract: | As Moléculas de orientação repulsivas (RGM) são uma família de proteínas multifuncionais composta por RGMa, RGMb, HFE2 e RGMd. Estudos recentes demonstraram que a proteína RGMa está ligada ao desenvolvimento do tecido muscular esquelético, controlando o tamanho e o mecanismo de fusão dos mioblastos em miotúbulos multinucleados. A superexpressão de RGMa na célula provoca o aparecimento de células hipertróficas, e sua ausência provoca o aparecimento de células atróficas. O padrão esperado de expressão dessa molécula nas células musculares esqueléticas está na membrana plasmática, mas o encontramos no núcleo das células-tronco musculares – as células satélites. Neste trabalho, levantou-se a hipótese de que a presença da proteína RGMa no núcleo das células satélites é impulsionada pelo domínio intracelular do seu receptor canônico, a neogenina. Células satélites do extensor longo dos dedos C57 Black foram isoladas de camundongos adultos. Posteriormente, a expressão endógena da neogenina foi derrubada utilizando transfecção de siRNA. As expressões de RGMa e neogenina foram determinadas por microscopia de imunofluorescência. Os resultados principais confirmaram que a proteína RGMa estava localizada nos núcleos das células satélites, enquanto a neogenina poderia ser encontrada na região perinuclear. A microscopia confocal mostrou que os picos fluorescentes estão no meio da pilha z, concluindo que o RGMa é expresso nos núcleos das células satélites. Em seguida, o western blot revelou que o RGMa37 ancorado é possivelmente o isotipo presente nos núcleos das células e o N-RGMa está no citoplasma da célula satélite. Além disso, uma nova banda de 50kDa pode ser expressa juntamente com RGMa37. Ao usar o siRNA-Neo1, a expressão da neogenina foi silenciada e o RGMa não pôde mais ser encontrado nos núcleos das células, sugerindo que a neogenina é o transportador para a nova expressão do RGMa nos núcleos das células satélites. A presença de RGMa-Neogenina no núcleo das células musculares esqueléticas implicará em possíveis novas funções biológicas para esta via de sinalização, que até o momento não foi descrita. |
