Resumen: | The central complex (CX) in the brain of insects is a highly conserved group of midline-spanning neuropils consisting of the upper and lower division of the central body,the protocerebral bridge, and the paired noduli. These neuropils are the substratefor a number of behaviors, most prominently goal-oriented locomotion. Honeybeeshave been a model organism for sky-compass orientation for more than 70 years, butthere is still very limited knowledge about the structure and function of their CX. Toadvance and facilitate research on this brain area, we created a high-resolution three-dimensional atlas of the honeybee’s CX and associated neuropils, including the poste-rior optic tubercles, the bulbs, and the anterior optic tubercles. To this end, we devel-oped a modified version of the iterative shape averaging technique, which allowed us toachieve high volumetric accuracy of the neuropil models. For a finer definition of spatiallocations within the central body, we defined layers based on immunostaining againstthe neuropeptides locustatachykinin, FMRFamide, gastrin/cholecystokinin, and allato-statin and included them into the atlas by elastic registration. Our honeybee CX atlasprovides a platform for future neuroanatomical work. |
Abstract: | O complexo central (CX) no cérebro de insetos é um grupo altamente conservado de neuropilos que abrangem a linha média, consistindo na divisão superior e inferior do corpo central, na ponte protocerebral e nos nódulos pareados. Esses neuropilos são o substrato para uma série de comportamentos, mais proeminentemente a locomoção orientada a objetivos. As abelhas têm sido um organismo modelo para orientação de bússola celeste por mais de 70 anos, mas ainda há conhecimento muito limitado sobre a estrutura e função de seu CX. Para avançar e facilitar a pesquisa sobre essa área do cérebro, criamos um atlas tridimensional de alta resolução do CX da abelha e neuropilos associados, incluindo os tubérculos ópticos posteriores, os bulbos e os tubérculos ópticos anteriores. Para esse fim, desenvolvemos uma versão modificada da técnica de média de forma iterativa, que nos permitiu atingir alta precisão volumétrica dos modelos de neuropilos. Para uma definição mais fina de localizações espaciais dentro do corpo central, definimos camadas com base na imunocoloração contra os neuropeptídeos locustaquicinina, FMRFamida, gastrina/colecistocinina e alatostatina e os incluímos no atlas por registro elástico. Nosso atlas CX de abelhas fornece uma plataforma para futuros trabalhos neuroanatômicos. |
