dc.description.abstract | O consumo crônico de etanol é uma das principais causas de mortalidade em todo o mundo, com maior risco de desenvolver infecções pulmonares, incluindo infecções por Aspergillus. Os mecanismos subjacentes ao aumento da suscetibilidade a infecções são pouco compreendidos. O consumo crônico de etanol induziu aumento das taxas de mortalidade, maior carga de Aspergillus fumigatus e redução do recrutamento de neutrófilos nas vias aéreas. A microscopia intravital mostrou diminuição na adesão leucocitária e rolamento após o consumo de etanol. Além disso, a ativação de neutrófilos diminuída e os níveis aumentados de CXCL1 sérico em camundongos alimentados com etanol induziram a internalização do receptor CXCR2 em neutrófilos circulantes. Os neutrófilos derivados da medula óssea de camundongos alimentados com etanol apresentaram menor depuração fúngica e produção defeituosa de espécies reativas de oxigênio. Em conjunto, os resultados mostraram que o etanol afeta as funções de ativação, recrutamento, fagocitose e morte de neutrófilos, causando suscetibilidade à infecção pulmonar por A. fumigatus. dc.description.resumo | Chronic ethanol consumption is a leading cause of mortality worldwide, with higher risks to develop pulmonary infections, including Aspergillus infections. Mechanisms underlying increased susceptibility to infections are poorly understood. Chronic ethanol consumption induced increased mortality rates, higher Aspergillus fumigatus burden and reduced neutrophil recruitment into the airways. Intravital microscopy showed decrease in leukocyte adhesion and rolling after ethanol consumption. Moreover, downregulated neutrophil activation and increased levels of serum CXCL1 in ethanol-fed mice induced internalization of CXCR2 receptor in circulating neutrophils. Bone marrow-derived neutrophils from ethanol-fed mice showed lower fungal clearance and defective reactive oxygen species production. Taken together, results showed that ethanol affects activation, recruitment, phagocytosis and killing functions of neutrophils, causing susceptibility to pulmonary A. fumigatus infection. This study establishes a new paradigm in innate immune response in chronic ethanol consumers. | pt_BR |
