Abstract: | A microbiota intestinal humana (MI) consiste em um grupo de microorganismos que residem no trato digestivo, em uma relação simbiótica com o hospedeiro. Existe um eixo intestino-cérebro, um sistema de comunicação bidirecional neuro-humoral, que integra o intestino e o cérebro do hospedeiro. O desequilíbrio da MI (disbiose) pode aumentar a permeabilidade da barreira intestinal e comprometer as vias de comunicação desse eixo. Disfunções no eixo microbiota-intestino-cérebro têm sido associadas a doenças do sistema nervoso central, como a doença de Alzheimer (DA) e outras doenças neurodegenerativas associadas ao envelhecimento. A DA é caracterizada clinicamente por um declínio progressivo e gradual na função cognitiva e neuropatologicamente pela presença de depósitos de placas extracelulares de peptídeo beta amiloide (Aβ) e por emaranhados neurofibrilares de proteína tau. Estudos em humanos e com modelos animais indicam que dietas com uma alimentação balanceada podem proporcionar benefícios, restabelecendo a eubiose intestinal. Essas descobertas fornecem evidências de compostos e de nutrientes potencialmente viáveis para a prevenção e tratamento da DA. O objetivo desta revisão literária é resumir e discutir as descobertas atuais que podem elucidar o papel da dieta na MI, a relação com o eixo intestino-cérebro e com as funções cognitivas. O entendimento desses mecanismos pode estimular a adoção de estratégias de prevenção e terapêuticas para a DA. |
Abstract: | The human intestinal microbiota (IM) consists in a group of microorganisms that live in the digestive tract, in a symbiotic relationship with the host. There is a gut-brain-axis, a bidirectional neuro-humoral communication system, that integrates the host gut and brain activities. IM imbalance (dysbiosis) can increase the permeability of the intestinal barrier and it affects the communication routes of this axis. Dysfunctions in the microbiota-gut-brain axis have been associated with diseases of the nervous system, such as Alzheimer's disease (AD) and other neurodegenerative diseases associated with aging. AD is characterized clinically by a progressive and gradual decrease in cognitive function and neuropathologically by the presence of extracellular plaque deposits of the β-amyloid peptide (Aβ) and the neurofibrillary tangles of the protein tau. Studies with animal models and human indicate that balanced diet can provide benefits, restoring intestinal eubiosis. These findings provide evidence of potentially viable compounds and nutrients for the prevention and treatment of AD. The purpose of this literature review is to summarize and discuss the current findings that may elucidate the role of diet in IM, the relationship with the gut-brain axis and cognitive functions. The understanding of these mechanisms can encourage the adoption of preventive and therapeutic strategies for AD. |
