Abstract: | Immunosuppression is a well-established risk factor for Visceral Leishmaniasis. Post-immunosuppression leishmaniasis is characterized by an increase of parasite burden, hematopoietic disorders and unusual clinical manifestations. Although there are many reports on bone marrow findings in VL, less is known about the relationship between parasite dynamics in this organ and the function of either hematopoietic stem cells and progenitor cells themselves. In the present study, we tackle these issues using a new approach of infecting human stem cells derived from bone marrow with L. infantum. Using this strategy, we show that human hematopoietic stem cells (hHSC) are able to phagocytize L. infantum promastigotes and release modulatory and pro-inflammatory cytokines, mainly TNF-α. Our results demonstrated that L. infantum infection in vitro enhances hematopoiesis, favoring the development of erythrocitic lineage through a mechanism yet unknown. Moreover, we found that L. infantum infection alters the phenotypic profile of the hematopoietic progeny; modifying the surface markers expression of differentiated cells. Thus, our study represents a rare opportunity to monitor the in vitro differentiation of human stem cells experimentally infected by L. infantum to better understand the consequences of the infection on phenotypic and functional profile of the cell progeny. |
Abstract: | A imunossupressão é um fator de risco bem estabelecido para a Leishmaniose Visceral. A leishmaniose pós-imunossupressão é caracterizada por um aumento da carga parasitária, distúrbios hematopoiéticos e manifestações clínicas incomuns. Embora existam muitos relatos sobre os achados da medula óssea na LV, pouco se sabe sobre a relação entre a dinâmica parasitária nesse órgão e a função das células-tronco hematopoiéticas e das próprias células progenitoras. No presente estudo, abordamos essas questões usando uma nova abordagem de infecção de células-tronco humanas derivadas da medula óssea com L. infantum. Usando esta estratégia, mostramos que células-tronco hematopoiéticas humanas (hHSC) são capazes de fagocitar promastigotas de L. infantum e liberar citocinas moduladoras e pró-inflamatórias, principalmente TNF-α. Nossos resultados demonstraram que a infecção in vitro por L. infantum potencializa a hematopoiese, favorecendo o desenvolvimento da linhagem eritrocítica através de um mecanismo ainda desconhecido. Além disso, descobrimos que a infecção por L. infantum altera o perfil fenotípico da progênie hematopoiética; modificar a expressão de marcadores de superfície de células diferenciadas. Assim, nosso estudo representa uma rara oportunidade de monitorar a diferenciação in vitro de células-tronco humanas experimentalmente infectadas por L. infantum para melhor compreender as consequências da infecção no perfil fenotípico e funcional da progênie celular. |
