Abstract: | O naftaleno é um dos hidrocarbonetos aromáticos policíclicos mais comumente encontrado no ambiente. A degradação do naftaleno por Pseudomonas putida G7 se deve à presença do plasmídeo NAH7, que codifica as enzimas da via superior envolvidas na conversão do naftaleno a salicilato, e às enzimas da via inferior envolvidas na conversão do salicilato a piruvato e acetato. Este trabalho buscou o entendimento das características estruturais e reacionais das enzimas NahC, NahF, NahI e NahG desta via. Em relação a NahC, este trabalho versou sobre estudos computacionais sobre o sítio ativo da NahC para avaliar a interação do centro metálico de ferro com os substratos 1,2-dihidroxinaftaleno e dioxigênio. Os estados de spin das estruturas que melhor descreveram os sistemas foram os estados singleto e tripleto para o complexo NahC-1,2-dihidroxinaftaleno e NahC-1,2-dihidroxinaftaleno-dioxigênio, respectivamente. Cálculos ONIOM mostraram que os resíduos H200, H246 e Y256, presentes na segunda esfera de coordenação do metal, são potencialmente importantes para o mecanismo de catálise da NahC. Estudos de inibição do salicilato nas enzimas NahF e NahI foram realizados para melhor entendimento da participação dessas duas aldeído desidrogenases na via metabólica da Pseudomonas putida G7. O perfil de inibição mostrou um tipo misto de inibição em que o inibidor se liga tanto ao sítio de ligação do substrato quanto do cofator, sendo que a NahI exibe uma maior sensibilidade a essa inibição em relação a NahF. Em relação a NahG, o trabalho buscou um entendimento da dinâmica da alça proteica que afeta a catálise enzimática. Para isso, foram explorados estudos com cofatores truncados que mostraram mudanças nos valores de Kd e kcat consistentes com alterações na energia de interação do cofator FAD. Experimentos de mutação sítio dirigida na alça de ligação do FAD foram feitos, para avaliar resíduos importantes tanto na ligação do FAD quanto na catálise da enzima NahG. |
Abstract: | Naphthalene is one of the most common polycyclic aromatic hydrocarbons in the environment. The degradation of naphthalene by Pseudomonas putida G7 is due to the presence of the NAH7 plasmid encoding the higher pathway enzymes involved in the conversion of naphthalene to salicylate and the lower pathway enzymes involved in the conversion of salicylate to pyruvate and acetate. This work aimed to understand the structural and reactional characteristics of NahC, NahF, NahI, and NahG of this pathway. In relation to NahC, this work focused on studies on the NahC active site to evaluate the interaction of the iron center with the 1,2-dihydroxynaphthalene and dioxygen substrates. The spin states of the structures that best described the systems were the singlet and triplet state for the NahC-1,2-dihydroxynaphthalene and NahC-1,2-dihydroxynaphthalene-dioxygen complex, respectively. ONIOM calculations showed that residues H200, H246 and Y256 in the second coordination sphere of the metal are potentially important for the catalysis mechanism of NahC. Salicylate inhibition studies on the NahF and NahI enzymes were performed for a better understanding of the role of these two aldehyde dehydrogenases in the metabolic pathway of Pseudomonas putida G7. The inhibition profile showed a mixed type of inhibition wherein the inhibitor binds to both the substrate binding site and the cofactor, for which NahI exhibited a greater sensitivity to inhibition regard NahF. In relation to NahG, the work aimed to understand the protein dynamics and interactions with FAD affecting the enzymatic catalysis. The studies with truncated cofactors showed changes in Kd and kcat values consistent with changes in the interaction energy of the FAD cofactor. Site-directed mutation experiments in the FAD binding loop were performed to evaluate the role of important residues in both FAD binding and NahG enzyme catalysis. |
