Abstract: | O reparo de feridas cutâneas é um processo biológico caracterizado por uma sequência estereotipada de eventos, que se sobrepõem de forma contínua e temporal e envolvem interações entre vários tipos celulares. Dentre tais eventos está a fase inflamatória, caracterizada por um proeminente infiltrado de diferentes tipos de leucócitos, dentre eles os linfócitos. Estas células, além de pouco estudadas no reparo comparadas a neutrófilos e macrófagos, participam de um fenômeno imunológico chamado tolerância oral, o qual é desencadeado pela ingestão de proteínas durante a alimentação. Nosso grupo de pesquisa mostrou que a reexposição parenteral a proteínas da dieta (tolerógenos), concomitante a lesões cutâneas em camundongos, reduz o infiltrado inflamatório no leito das feridas e melhora a cicatrização. Sabe-se pouco ainda sobre os mecanismos desencadeados por essa reexposição, mas há indícios do envolvimento de linfócitos T. Para entender um pouco mais sobre este envolvimento, neste trabalho analisou-se o efeito da reexposição parenteral a tolerógenos da dieta sobre o reparo de lesões cutâneas em camundongos nude, os quais apresentam deficiência de linfócitos T, e em camundongos heterozigotos. Observou-se que a administração parenteral de proteínas toleradas promoveu modificações em aspectos inflamatórios e na deposição de matriz ao 7° dia pós-lesão, e no tecido cicatricial ao 40° dia pós-lesão. Essas modificações ocorreram tanto em camundongos nude quanto em heterozigotos, mas de forma mais branda do que se observa normalmente em animais WT. Estes resultados indicam que os efeitos da reexposição parenteral a tolerógenos sobre o reparo cutâneo ocorrem em camundongos deficientes em linfócitos T, ainda que de forma mais branda e com um perfil inflamatório diferente do usualmente observado. |
Abstract: | Skin wound healing is a biological process characterized by a stereotyped sequence of events, which overlap each other in a continuous temporal manner and involve interactions between several cellular types. The inflammatory phase is among these events, and it is marked by an intense infiltrate of diferent types of leukocytes, including lymphocytes. These cells have a less studied role at wound healing, compared to macrophages and neutrophils, but they are known to be involved at oral tolerance, an immunological event triggered by protein intake at feeding. Our research group showed that parenteral re-exposure to dietary proteins (tolerogens), concomitant to skin wounding at mice, reduces the inflammatory infiltrate at the wound bed and improves the wound repair. Little is known about the mechanisms triggered by this parenteral re-exposure, but it is known that T lymphocytes are involved. In order to understand a little more about T cell role, in this study the effects of tolerogen parenteral re-exposure over skin wound repair were analysed at nude mouse (FoxN1-/-), which presents a T cell deficiency, and at heterozygous (FoxN1+/-) mouse. It was observed that parenteral administration of tolerated proteins promoted modifications in inflammatory aspects and matrix deposition at the 7th day post-injury, and in tissue repair at the 40th day post-injury. These modifications occured at both nude and heterozygous mouse, but at a smaller degree then it is normally observed at WT mouse. These results indicate that the effects of parenteral reexposure to tolerogens over skin repair occurs in T lymphocyte deficient mice, although in a milder way and with a different inflammatory profile than usually observed. |
