Abstract: | A capsaicina, um composto pungente das pimentas é descrito como um anti-inflamatório potente, além de ter propriedades antioxidantes e antimicrobianas, modulando o sistema imune e a microbiota. Nosso objetivo foi avaliar o efeito do uso tópico de capsaicina na melhora da inflamação e função da barreira epitelial em colite experimental induzida por sulfato dextrano de sódio (DSS). Para tal, camundongos C57/BL6 controles ou com colite induzida DSS receberam ou não creme de capsaicina a 0,075% por 6 dias. Nossos resultados mostraram pela primeira vez, que o creme de capsaicina diminuiu a perda de peso, encurtamento do cólon e a diarreia, típicos da colite. A farmacocinética da capsaicina marcada com tecnécio radioativo mostrou que o creme de capsaicina aplicado no dorso do animal alcança o cólon e pode atuar diretamente no local. O uso do creme de capsaicina levou à melhora da função de barreira epitelial, com preservação de proteínas das junções firme, aderente e dos desmossomos. Nos linfonodos cecais dos animais tratados foi vista uma menor frequência de células T CD4+ (RORT, LAP e Treg). Observamos também que o grupo tratado com creme de capsaicina melhorou o perfil de ácidos graxos de cadeia curta no ceco. Por fim, observamos maior expressão do receptor de capsaicina TRPV1 nos grupos recebendo creme de capsaicina. Em conclusão, nossos resultados sugerem que a capsaicina em administração tópica pode ser um adjuvante promissor no tratamento de colite, reduzindo a inflamação e melhorando a função de barreira intestinal. |
Abstract: | Capsaicin, a pungent compound of peppers is described as a potent anti-inflammatory agent, besides having antioxidant and antimicrobial properties, modulating the immune system and microbiota. Our objective was to evaluate the effect of topical use of capsaicin on the improvement of inflammation and epithelial barrier function in experimental colitis induced by dextran sodium sulphate (DSS). For this, C57/BL6 mice, healthy or with DSS induced colitis received or not capsaicin cream at 0.075% for 6 days. Our results showed for the first time that capsaicin cream reduced weight loss, colon shortening and diarrhea, all typical signs of colitis. The pharmacokinetics of radioactive technetium labelled capsaicin shows that the capsaicin cream applied to the back of the animal reaches the colon and can act directly on that site. The use of capsaicin cream led to the improvement of epithelial barrier function, preserving proteins of thigh, adherent junctions and desmossomes. A lower frequency of CD4 T cells (RORT, LAP e Treg) was seen in the cecal lymph nodes of the capsaicin treated animals. We also observed that the group treated with capsaicin cream improved the profile of short-chain fatty acids in the cecum. Finally, we observed higher expression of the capsaicin receptor TRPV1 in the groups receiving capsaicin cream. In conclusion, our results suggest that topical administration of capsaicin may be a promising adjuvant in the treatment of colitis, reducing inflammation and improving intestinal barrier function. |
