Abstract: | No testículo de suínos é possível identificar dois tipos morfologicamente distintos de células de Leydig, os quais encontram-se preferencialmente distribuídos na região intermediária (ID) e na região periférica (RP) do parênquima testicular. Assim, células hipertrofiadas predominam em ID e células com menor volume individual são frequentes em RP. O presente estudo teve como principal objetivo investigar características fenotípicas que pudessem auxiliar na compreensão das possíveis diferenças funcionais entre as duas populações. Assim, em testículos de suínos pós-púberes observamos que as células de Leydig presentes em RP apresentaram menor intensidade de marcação para o ERα e concomitante aumento no índice de proliferação celular, não sendo verificadas alterações quanto ao padrão de imunorreatividade para AR e LHR. A fim de avaliar a capacidade esteroidogênica das diferentes regiões, realizamos um estudo in vivo utilizando o modelo de xenoenxerto de fragmentos de testículos de doadores suínos jovens. Desse modo pudemos observar uma tendência quanto à ID ser mais eficiente para a síntese de testosterona. Ademais, em RP, a densidade de macrófagos apresentou forte tendência a ser maior. Diante desses achados, sugerimos que na população de células de Leydig em RP predominam células progenitoras e imaturas da linhagem adulta, enquanto em ID estão presentes as células maduras. Contudo, mais estudos serão desenvolvidos para melhor compreendermos as interações celulares nestas regiões distintas do parênquima testicular. |
Abstract: | Two morphologically distinct Leydig cells types are recognized within the interstitial compartment of pigs testes. Moreover, these cells seem to present a preferential distribution, since the hypertrophied cell type is observed in the intermediate region (ID), and those cells with small individual volume are more frequent in the peripheral region of testis parenchyma. Therefore, the present study aimed to phenotypically characterize these two cell populations, in order to better understand the functional differences they may present. In this regard, in testes from postpubertal pigs, we observed that Leydig cells in RP presented a weak intensity of ERα labeling, whereas the proliferation index found for this population was higher. No differences regarding AR and LHR immunoreactivity were observed. Additionally, we evaluated the steroidogenic function in ID and RP through the xenografting approach that enabled to assess in vivo the synthesis of androgens. Indeed, ID presented a clear trend and the recipient showed high levels of testosterone. On the other hand, macrophages were more observed in RP. These findings suggest that Leydig cells in RP are mainly progenitors and immature types, whereas ID maintain the mature type. However, further studies are being developed so to better comprehend the cells’ interactions and how they modulate the different regions of testis parenchyma. |
