Abstract: | Em busca de entender os efeitos da migração larval, e o processo infeccioso de maneira interdisciplinar, o objetivo do presente estudo foi avaliar os impactos da infecção por Toxocara canis durante a fase subaguda e crônica da infecção em modelo murino BALB/c, considerando parâmetros parasitológicos, psicocomportamentais, imunopatológicos e histopatológicos da infecção. Para cumprir com o objetivo, os camundongos BALB/c foram divididos de acordo com a quantidade de ovos inoculados, grupo G100, inoculado com 100 ovos embrionados e grupo G1000, inoculado com 1.000 ovos embrionados. Como principais resultados obtidos, se destaca uma maior recuperação de larvas L3 no grupo G1000 em comparação ao grupo G100, com maior recuperação de larvas no tecido encefálico. Na avaliação psicobiológica do comportamento encontramos um comportamento tipo ansiogênico nos animais G100 e comportamento tipo ansiolítico no grupo G1000. As citocinas IL-6 e TNF- α, e a quimiocina CCL-17, marcadores esses pró-inflamatórias, serviram como marcadores secundários da infecção por Toxocara canis. Na avaliação histopatológica do tecido pulmonar, encontramos scores de inflamação aumentados nos animais do grupo G1000, eutanasiados tanto no tempo 14DPI como 42DPI. Já a histopatologia do tecido hepático, apresentou-se, no grupo G1000 eutanasiado no tempo 42DPI, níveis baixos de granuloma no tempo 42DPI. Enquanto a coloração usada para análise histopatológica encefálica, não detectou alterações fisiopatológicas de tecido nos animais infectados. Em conclusão, foram detectadas alterações histopatológicas e imunológicas no grupo de baixa (G100) e alta (G1000) dose de infecção por Toxocara canis. Quanto ao peso dos animais, encontramos ganhos de acordo com a normalidade. Porém, apenas no grupo G1000 foi possível recuperar larvas migratórias, principalmente no encéfalo. Além disso, encontramos diferenças quanto ao comportamento tipo ansioso, apresentando o grupo de baixa (G100) dose de infecção um comportamento tipo ansiolítico e o grupo de alta (G1000) dose de infecção um comportamento tipo ansiogênico, já o comportamento tipo depressivo foi encontrado na fase crônica dos animais G1000. |
Abstract: | In order to understand the effects of larval migration, and the process of infection, this study aimed to analyse the effects of T. canis infection in BALB/c mice during the acute and chronic phases of infection. This study specifically evaluated parasitological, psychobehavioral, immunopathological and histopathological parameters during the course of infection. BALB/c mice were grouped according to the amount of inoculated eggs, group G100, with 100 embryonated eggs, and group G1000 with 1,000 embryonated eggs. Results demonstrated a higher recovery of L3 larvae from the G1000 group compared to the G100 group, with the most larvae recovered from brain tissue. In the psychobiological evaluation of behaviour we found an anxiolytic-like behaviour in the G100 animals and anxiogenic-like behaviour in the G1000 group. The evaluation of inflammatory markers showed an increase in IL-6 concentrations in infected animals at 42DPI, as well as an increase in relation to TNF-α and CCL-17 concentrations in the G1000 group. As for the histopathological evaluation of lung tissue, we found increased inflammation scores in the animals of the G1000 group at both 14DPI and 42DPI. Additionally, the histopathology of the liver tissue showed low levels of granuloma in the G1000 group at 42DPI, while the chosen staining methods for brain histopathology showed no pathophysiological tissue changes in any of the analysed groups. In conclusion, histopathological and immunological changes were detected after low (G100) and high (G1000) doses of Toxocara canis infection. As for the weight of the animals, we found normal weight gains in the course of infection. However, only in the G1000 group it was possible to recover migrating larvae, mainly in the brain. In addition, we found differences regarding anxiety-like behavior, with the low (G100) infection-type group presenting an anxiolytic-like behavior and the high (G1000) infection-dose group presenting an anxiogenic-like behavior, whereas the depressive-like behavior was found in the G1000 group in the chronic phase. |
