Abstract: | Brucella abortus is the etiological agent of bovine brucellosis, a zoonotic disease that causes significant economic losses worldwide. The differential proteomic profile of bovine chorioallantoic membrane (CAM) explants at early stages of infection with B. abortus (0.5, 2, 4, and 8 h) was determined. Analysis of CAM explants at 0.5 and 4 h showed the highest differences between uninfected and infected CAM explants, and therefore were used for the Differential Gel Electrophoresis (DIGE). A total of 103 spots were present in only one experimental group and were selected for identification by mass spectrometry (MALDI/ToFToF). Proteins only identified in extracts of CAM explants infected with B. abortus were related to recognition of PAMPs by TLR, production of reactive oxygen species, intracellular trafficking, and inflammation. |
Abstract: | Brucella abortus é o agente etiológico da brucelose bovina, uma zoonose que causa perdas econômicas significativas em todo o mundo. O perfil proteômico diferencial de explantes de membrana corioalantóide bovina (CAM) em estágios iniciais de infecção com B. abortus (0,5, 2, 4 e 8 h) foi determinado. A análise de explantes CAM em 0,5 e 4 h mostrou as maiores diferenças entre explantes CAM não infectados e infectados e, portanto, foram usados para a Eletroforese Diferencial em Gel (DIGE). Um total de 103 spots estavam presentes em apenas um grupo experimental e foram selecionados para identificação por espectrometria de massa (MALDI/ToFToF). Proteínas identificadas apenas em extratos de explantes CAM infectados com B. abortus foram relacionadas ao reconhecimento de PAMPs por TLR, produção de espécies reativas de oxigênio, tráfego intracelular e inflamação. |
