Abstract: | Calcium (Ca 2+) is an important second messenger in charge of many critical processes in the central nervous system (CNS), including membrane excitability, neurotransmission, learning, memory, cell proliferation, and apoptosis. In this way, the voltage-gated calcium channels (VGCCs) act as a key supply for Ca2+ entry into the cytoplasm and organelles. Importantly, the dysregulation of these channels has been reported in many neurological diseases of young-onset, with associated genetic factors, such as migraine, multiple sclerosis, and Huntington’s disease. Notably, the literature has pointed to the role of N-type Ca2+ channels (NTCCs) in controlling a variety of processes, including pain, inflammation, and excitotoxicity. Moreover, several Ca2+ channel blockers that are used for therapeutic purposes have been shown to act on the N-type channels. Therefore, this review provides an overview of the NTCCs in neurological disorders focusing mainly on Huntington’s disease, multiple sclerosis, and migraine. It will discuss possible strategies to generate novel therapeutic strategies. |
Abstract: | O cálcio (Ca 2+) é um importante segundo mensageiro responsável por muitos processos críticos no sistema nervoso central (SNC), incluindo excitabilidade da membrana, neurotransmissão, aprendizagem, memória, proliferação celular e apoptose. Dessa forma, os canais de cálcio dependentes de voltagem (VGCCs) atuam como um suprimento chave para a entrada de Ca2+ no citoplasma e nas organelas. É importante ressaltar que a desregulação desses canais tem sido relatada em muitas doenças neurológicas de início precoce, com fatores genéticos associados, como enxaqueca, esclerose múltipla e doença de Huntington. Notavelmente, a literatura tem apontado para o papel dos canais de Ca2+ tipo N (NTCCs) no controle de uma variedade de processos, incluindo dor, inflamação e excitotoxicidade. Além disso, vários bloqueadores dos canais de Ca2+ utilizados para fins terapêuticos demonstraram atuar nos canais do tipo N. Portanto, esta revisão fornece uma visão geral dos NTCCs em distúrbios neurológicos, concentrando-se principalmente na doença de Huntington, esclerose múltipla e enxaqueca. Discutirá possíveis estratégias para gerar novas estratégias terapêuticas. |
