Abstract: | The role of suppressors of cytokine signaling (SOCS) in meningoencephalitis caused by Bovine herpesvirus 5 (BoHV-5) was evaluated by intracranial infection in C57BL/6 wild-type mice (WT) and SOCS2 deficient mice (SOCS2(-/-)). Both infected groups presented weight loss, ruffled fur and hunched posture. Additionally, infected SOCS2(-/-) mice showed swollen chamfer and progressive depression. Infected WT animals developed mild meningitis, characterized by infiltration of mononuclear cells. Moreover, viral DNA was detected in liver and lung from infected WT group. This group also showed elevated brain levels of IFN-γ, IL-10, CXCL1 and CCL5, when compared with non-infected WT animals. Brain inflammation was exacerbated in infected SOCS2(-/-) mice with widespread distribution of the virus and increased brain levels of TNF-α, IFN-γ, IL-10, IL-12, CXCL1 and CCL5, when compared with WT infected mice. Moreover, infected SOCS2 deficient mice exhibited reduced brain mRNA expression of IFNα and IFNβ and increased expression of mRNA of SOCS1, compared with infected WT mice. Taken together, our study provides an insight into the role of SOCS2 in modulating the immune response to BoHV-5 infection. |
Abstract: | O papel dos supressores de sinalização de citocinas (SOCS) na meningoencefalite causada pelo herpesvírus bovino 5 (BoHV-5) foi avaliado por infecção intracraniana em camundongos C57BL/6 do tipo selvagem (WT) e camundongos deficientes em SOCS2 (SOCS2(-/-)) . Ambos os grupos infectados apresentaram perda de peso, pelagem arrepiada e postura curvada. Além disso, camundongos SOCS2(-/-) infectados apresentaram chanfro inchado e depressão progressiva. Animais WT infectados desenvolveram meningite leve, caracterizada por infiltração de células mononucleares. Além disso, o DNA viral foi detectado no fígado e no pulmão do grupo WT infectado. Este grupo também apresentou níveis cerebrais elevados de IFN-γ, IL-10, CXCL1 e CCL5, quando comparado com animais WT não infectados. A inflamação cerebral foi exacerbada em camundongos SOCS2 (-/-) infectados com ampla distribuição do vírus e aumento dos níveis cerebrais de TNF-α, IFN-γ, IL-10, IL-12, CXCL1 e CCL5, quando comparado com camundongos infectados por WT . Além disso, camundongos infectados com deficiência de SOCS2 exibiram redução da expressão de mRNA cerebral de IFNα e IFNβ e aumento da expressão de mRNA de SOCS1, em comparação com camundongos WT infectados. Em conjunto, nosso estudo fornece uma visão sobre o papel do SOCS2 na modulação da resposta imune à infecção pelo BoHV-5. |
