Abstract: | O regime terapêutico para o tratamento da AIDS utilizando efavirenz, lamivudina e fumarato de tenofovir desoproxila tem demonstrado eficácia e segurança. Neste trabalho, um método analítico foi desenvolvido e validado para a quantificação simultânea de efavirenz (EFV), lamivudina (3TC) e fumarato de tenofovir desoproxila (TDF) em comprimidos de dose fixa combinada. A análise por cromatografia a líquido de alta eficiência foi realizada em coluna de fase reversa Sun Fire® C18 (250 mm x 4,6 mm; 5 μm) a 30 °C, com 20 μL de volume de injeção e detecção a 260 nm. A fase móvel, composta por metanol e solução tampão acetato pH 5,4, foi utilizada em vazão de 1,0 mL/min, sob gradiente. Foi obtida linearidade adequada na faixa de 66 a 198 μg/mL para EFV, 66 a 198 μg/mL para 3TC e 33 a 99 μg/mL para TDF. Os valores de desvio padrão relativo foram inferiores a 5% e recuperações médias no intervalo de 98% a 102%, confirmando a precisão e a exatidão do método, respectivamente. Além disso, o método desenvolvido mostrou-se seletivo e robusto, podendo ser aplicado com sucesso a análises rotineiras de controle de qualidade. |
Abstract: | The therapeutic regimen for the treatment of AIDs using efavirenz, lamivudine and tenofovir disoproxil fumarate has demonstrated efficacy and safety. In this work, an analytical method was developed and validated for simultaneous quantitation of efavirenz (EFV), lamivudine (3TC) and tenofovir disoproxil fumarate (TDF) in fixed-dose combination tablet. Analysis by high performance liquid chromatography was performed using a reversed phase column sun Fire® C18 (250 mm x 4,6 mm; 5 μm) at 30 °C, 20 μL injection volume and detection at 260 nm. The mobile phase, composed of acetate buffer pH 5.4 and methanol, was employed at 1.0 mL/min flow rate and gradient elution. Adequate linearity was obtained in the range from 66 to 198 μg/mL for EFV, 66 to 198 μg/mL from 3TC and 33 to 99 μg/mL for TDF. Relative standard deviation values were lower than 5% and mean recoveries were within 98% to 102%, confirming the precision and accuracy of the method, respectively. In addition, the developed method showed to be selective and robust and can be successfully applied for routine quality control analysis. |
