Abstract: | Malaria in pregnancy (MiP) induces intrauterine growth restriction (IUGR) and pre-term labour (PTL). However, its effects on yolk sac morphology and function arelargely unexplored. We hypothesized that MiP modifies yolk sac morphology andefflux transport potential by modulating ABC efflux transporters. C57BL/6 mice in-jected with Plasmodium berghei ANKA (5 × 10 5 infected erythrocytes) at gestationalday (GD) 13.5 were subjected to yolk sac membrane harvesting at GD 18.5 for his-tology, qPCR and immunohistochemistry. MiP did not alter the volumetric propor-tion of the yolk sac's histological components. However, it increased levels of Abcb1amRNA (encoding P-glycoprotein) and macrophage migration inhibitory factor (Mifchemokine), while decreasing Abcg1 (P < 0.05); without altering Abca1, Abcb1b, Abcg2,Snat1, Snat2, interleukin (Il)-1β and C-C Motif chemokine ligand 2 ( Ccl2). Transcriptsof Il-6, chemokine (C-X-C motif) ligand 1 (Cxcl1), Glut1 and Snat4 were not detectible.ABCA1, ABCG1, breast cancer resistance protein (BCRP) and P-gp were primarilyimmunolocalized to the cell membranes and cytoplasm of endodermic epitheliumbut also in the mesothelium and in the endothelium of mesodermic blood vessels.Intensity of P-gp labelling was stronger in both endodermic epithelium and meso-thelium, whereas ABCA1 labelling increased in the endothelium of the mesodermicblood vessels. The presence of ABC transporters in the yolk sac wall suggests thatthis fetal membrane acts as an important protective gestational barrier. Changes in ABCA1 and P-gp in MiP may alter the biodistribution of toxic substances, xenobiotics,nutrients and immunological factors within the fetal compartment and participate inthe pathogenesis of malaria-induced IUGR and PTL. |
Abstract: | A malária na gravidez (MiP) induz restrição de crescimento intrauterino (RCIU) e trabalho de parto prematuro (PTL). No entanto, seus efeitos na morfologia e função do saco vitelino são amplamente inexplorados. Nossa hipótese é que o MiP modifica a morfologia do saco vitelino e o potencial de transporte de efluxo modulando os transportadores de efluxo ABC. Camundongos C57BL/6 injetados com Plasmodium berghei ANKA (5 × 105 eritrócitos infectados) no dia gestacional (GD) 13,5 foram submetidos à colheita da membrana do saco vitelino em GD 18,5 para histologia, qPCR e imuno-histoquímica. A MiP não alterou a proporção volumétrica dos componentes histológicos do saco vitelino. No entanto, aumentou os níveis de Abcb1amRNA (que codifica a glicoproteína P) e do fator inibidor da migração de macrófagos (Mifchemokine), enquanto diminuiu Abcg1 (P <0,05); sem alterar Abca1, Abcb1b, Abcg2, Snat1, Snat2, interleucina (Il) -1β e ligante de quimiocina CC Motif 2 (Ccl2). Transcrições de Il-6, ligante 1 de quimiocina (motivo C-X-C) (Cxcl1), Glut1 e Snat4 não foram detectáveis. ABCA1, ABCG1, proteína de resistência ao câncer de mama (BCRP) e P-gp foram principalmente imunolocalizados nas membranas celulares e citoplasma do epitélio endodérmico, mas também no mesotélio e no endotélio dos vasos sanguíneos mesodérmicos. A intensidade da marcação da P-gp foi mais forte tanto no epitélio endodérmico quanto no mesotélio, enquanto a marcação ABCA1 aumentou no endotélio dos vasos sanguíneos mesodérmicos. A presença de transportadores ABC na parede do saco vitelino sugere que esta membrana fetal atua como uma importante barreira protetora gestacional. Alterações no ABCA1 e na P-gp na MiP podem alterar a biodistribuição de substâncias tóxicas, xenobióticos, nutrientes e fatores imunológicos dentro do compartimento fetal e participar na patogênese da RCIU e PTL induzidas pela malária. |
