dc.description.abstract | A morte celular desempenha um papel fundamental na montagem da imunidade protetora e patogênica. A etose é um mecanismo de morte celular definido pela liberação de armadilhas extracelulares (ETs), que pode promover inflamação e exercer atividade microbicida. Embora a etose esteja frequentemente associada com células inatas, estudos recentes mostraram que células B e células T CD4+ podem liberar ETs. Aqui investigamos se as células T CD8+ também podem liberar ETs, o que pode estar relacionado a citotoxicidade e patologia tecidual. Para esses fins, primeiro empregamos um sistema in vitro estimulação de células T CD8+ humanas isoladas de voluntários saudáveis com anti-CD3/antiCD28. Usando vídeo de intervalo de tempo, microscopia confocal e eletrônica, demonstramos que células T CD8 + humanas liberam ETs após estimulação (aqui LETs – linfócitos extracelulares armadilhas), que apresentam morfologia e características funcionais únicas. LETs derivados de células T CD8+ formam longas cadeias que co-localizam com CD107a, um marcador de vesículas contendo grânulos citotóxicos. Além disso, essas estruturas conectam a célula liberadora de LET para outras células vizinhas, muitas vezes resultando em morte celular. Depois de demonstrar a liberação de LETs por células T CD8+ humanas in vitro, passamos a estudar a ocorrência de células derivadas de CD8 LETs em um ambiente de doença humana. Assim, avaliamos a ocorrência de LETs derivados de CD8 em lesões de pacientes com leishmaniose tegumentar humana, onde as células T CD8+ desempenham um papel papel fundamental na mediação da patologia. Além disso, avaliamos a associação desses estruturas com a intensidade do infiltrado inflamatório em cutâneo precoce e tardio, como bem como em lesões de leishmaniose mucosa. Demonstramos que a progressão e a gravidade de formas debilitantes e mutilantes de leishmaniose tegumentar humana estão associadas à frequência de células T CD8+ na etose, bem como à ocorrência de células T CD8+ derivadas LETs carregando vesículas CD107a+ nas lesões. Propomos que LETs derivados de células T CD8+ pode servir como uma ferramenta para entregar vesículas citotóxicas a células-alvo distantes, fornecendo insights sobre mecanismos de patologia mediada por células T CD8+. | pt_BR |
dc.description.resumo | Cell death plays a fundamental role in mounting protective and pathogenic immunity. Etosis is a cell death mechanism defined by the release of extracellular traps (ETs), which can foster inflammation and exert microbicidal activity. While etosis is often associated with innate cells, recent studies showed that B cells and CD4+ T cells can release ETs. Here we investigate whether CD8+ T cells can also release ETs, which might be related to cytotoxicity and tissue pathology. To these ends, we first employed an in vitro system stimulating human CD8+ T cells isolated from healthy volunteers with anti-CD3/antiCD28. Using time-frame video, confocal and electron microscopy, we demonstrate that human CD8+ T cells release ETs upon stimulation (herein LETs – lymphocyte extracellular traps), which display unique morphology and functional characteristics. CD8+ T cellderived LETs form long strands that co-localize with CD107a, a marker of vesicles containing cytotoxic granules. In addition, these structures connect the LET-releasing cell to other neighboring cells, often resulting in cell death. After demonstrating the release of LETs by human CD8+ T cells in vitro, we went on to study the occurrence of CD8-derived LETs in a human disease setting. Thus, we evaluated the occurrence of CD8-derived LETs in lesions from patients with human tegumentary leishmaniasis, where CD8+ T cells play a key role in mediating pathology. In addition, we evaluated the association of these structures with the intensity of the inflammatory infiltrate in early and late cutaneous, as well as in mucosal leishmaniasis lesions. We demonstrated that progression and severity of debilitating and mutilating forms of human tegumentary leishmaniasis are associated with the frequency of CD8+ T cells in etosis, as well as the occurrence of CD8-derived LETs carrying CD107a+ vesicles in the lesions. We propose that CD8+ T cell derived LETs may serve as a tool for delivering cytotoxic vesicles to distant target cells, providing insights into mechanisms of CD8+ T cell mediated pathology. | pt_BR |
