Abstract: | Visceral leishmaniasis is a chronic disease that affects humans and dogs as well. Dogs, the domestic reservoir of Leishmania, play a central role in the transmission of visceral leishmaniasis, the most severe form of this disease. Neutrophils are the most abundant leukocytes in blood and interact with the parasite after infection. Here, we evaluate the effector properties of neutrophils from healthy and naturally Leishmania infantum-infected dogs. Our results showed that the parasite induced neutrophil extracellular trap (NET) release from neutrophils in both groups. Additionally, phagocytosis and NETs contributed differently to parasite killing by neutrophils from healthy and infected animals, and IFN-γ, IL-8, IL-4 and TNF-α production by neutrophils from both groups were differentially modulated by the parasite. Our results contribute to a better understanding of the complex role played by neutrophils in canine visceral leishmaniasis, which may favor the development of more effective therapies. |
Abstract: | A leishmaniose visceral é uma doença crônica que afeta também humanos e cães. Os cães, reservatório doméstico da Leishmania, desempenham um papel central na transmissão da leishmaniose visceral, a forma mais grave desta doença. Os neutrófilos são os leucócitos mais abundantes no sangue e interagem com o parasita após a infecção. Aqui, avaliamos as propriedades efetoras dos neutrófilos de cães saudáveis e naturalmente infectados por Leishmania infantum. Nossos resultados mostraram que o parasita induziu a liberação de armadilha extracelular de neutrófilos (NET) dos neutrófilos em ambos os grupos. Além disso, a fagocitose e os NETs contribuíram diferentemente para a morte do parasita pelos neutrófilos de animais saudáveis e infectados, e a produção de IFN-γ, IL-8, IL-4 e TNF-α pelos neutrófilos de ambos os grupos foi modulada diferencialmente pelo parasita. Nossos resultados contribuem para uma melhor compreensão do complexo papel desempenhado pelos neutrófilos na leishmaniose visceral canina, o que pode favorecer o desenvolvimento de terapias mais eficazes. |
