| Abstract: | We describe for the first time the karyotype of Ololygon machadoi (Amphibia, Anura, Hylidae) collected in southeastern Brazil. The chromosome preparations were obtained from cultured fibroblasts and this population presented a diploid number 2n = 24, a fundamental number FN = 48, and a karyotype that included eight metacentric, six submetacentric, and ten subtelocentric chromosomes (karyotypic formula 8m+6sm+10st). The CBG-banding patterns revealed constitutive heterochromatin in the pericentromeric regions of all chromosomes, and silver-staining showed that the nucleolar organizer regions (Ag-NORs) are located on the short arms of chromosome pair 6. Cytogenetic information of O. machadoi will contribute to future studies of this genus and the cell culture protocol for obtaining chromosome preparations will be useful for other amphibians. |
| Abstract: | Descrevemos pela primeira vez o cariótipo de Ololygon machadoi (Amphibia, Anura, Hylidae) coletado no sudeste do Brasil. As preparações cromossômicas foram obtidas de fibroblastos cultivados e esta população apresentou número diplóide 2n = 24, número fundamental FN = 48 e cariótipo que incluía oito cromossomos metacêntricos, seis submetacêntricos e dez subtelocêntricos (fórmula cariotípica 8m+6sm+10st) . Os padrões de bandeamento CBG revelaram heterocromatina constitutiva nas regiões pericentroméricas de todos os cromossomos, e a coloração com prata mostrou que as regiões organizadoras de nucléolos (Ag-NORs) estão localizadas nos braços curtos do par cromossômico 6. Informações citogenéticas de O. machadoi contribuirão para estudos futuros deste gênero e o protocolo de cultura celular para obtenção de preparações cromossômicas será útil para outros anfíbios. |
