Abstract: | O envolvimento dos clostrídios como agentes causadores de diarreia em leitões neonatos tem crescido no mundo todo. No Brasil, são escassos os relatos da ocorrência de Clostridium difficile e Clostridium perfringens tipos A e C como causadores de diarreia em leitões, devido à ausência de diagnóstico laboratorial destes agentes. Diante deste quadro, os objetivos desse estudo foram determinar a frequência dos clostrídios entéricos que acometem leitões neonatos com até sete dias de vida, em granjas comerciais acima de 500 matrizes, localizadas na região do Triângulo Mineiro e Alto Paranaíba, no estado de Minas Gerais, Brasil; e desenvolver e avaliar uma vacina polivalente, contendo os toxóides alfa de C. perfringens tipo A, beta de C. perfringens tipo C e A/B de C. difficile. Dos 60 animais utilizados neste trabalho, em 12 (20%) foi possível isolar e genotipificar C. difficile a partir das fezes, sendo cinco (41,7%) do grupo diarreico e sete (58,3%) do grupo não diarreico. Na interpretação dos dados da detecção qualitativa das toxinas A/B de C. difficile pelo ELISA, observou-se que dos 60 animais analisados em 10 (16,7%) foi possível a identificação das toxinas. Destes 10 leitões, sete (70%) eram do grupo diarreico e os outros três (30%) do grupo não diarreico. Na necropsia, 13 leitões (21,67%) apresentaram edema de mesocólon, principal alteração macroscópica associada com a infecção por C. difficile. Dos 13 animais que apresentavam edema de mesocólon, oito (61,54%) eram do grupo diarreico e os outros cinco (38,46%) do grupo não diarreico. À microscopia, 16 animais apresentaram lesões histopatológicas, caracterizadas por lesões inflamatórias. Destes 16 leitões, sete (43,75%) apresentavam o quadro clínico de diarreia e outros nove (56,25%) eram não diarreicos. Dos 60 animais utilizados neste trabalho, em 50 (83,3%) foi possível isolar colônias características de C. perfringens a partir das fezes. Com a tipificação destas amostras, observou-se que todas eram C. perfringens tipo A. Destas 50 amostras de C. perfringens tipo A, 23 (46%) foram isoladas de animais com diarreia e 27 (54%) isoladas de animais aparentemente sadios. Em 37 (74%) amostras de C. perfringens tipo A isoladas, foi detectada a presença do gene cpb2, codificador da toxina beta-2. O resultado do seqüenciamento do gene cpb2 revelou que apenas 10 amostras, oito do grupo não diarreico e duas do grupo diarreico, apresentavam uma mutação no gene cpb2+ que gerava uma proteína truncada sem ação efetiva. As outras 21 amostras, 12 do grupo diarreico e nove do grupo não diarreico, não apresentavam a mutação, permitindo assim a tradução normal da toxina beta-2. Não foi isolada nenhuma amostra de C. perfringens tipo C. A concentração inibitória mínima (CIM) estabelecida frente às amostras de C. perfringens tipo A demonstrou que amoxicilina, ceftiofur e a narasina, foram os antimicrobianos que apresentaram a melhor atividade inibitória. A vacina experimental polivalente contendo os toxóides alfa recombinante, beta recombinante e A/B nativo foi eficiente em estimular títulos de anticorpos neutralizantes nos animais estudados, demonstrando poder ser uma importante ferramenta para o controle das diarreias causadas por clostrídios na maternidade |
Abstract: | The involvement of clostridia as agents causing diarrhea in neonatal piglets has grown worldwide. In Brazil there are few reports of the occurrence of Clostridium difficile, Clostridium perfringens types A and C causing diarrhea in piglets due to the lack of laboratory diagnosis of these agents. Given this situation, the aims of this study were to determine the frequency of enteric clostridia affecting newborn piglets with up to seven days, on commercial farms over 500 sows, located in the Triangulo Mineiro and Alto Paranaíba, state of Minas Gerais, Brazil; and develop and evaluate a polyvalent vaccine containing the alpha toxoid of C. perfringens type A, beta toxoid of C. perfringens type C and A/B toxoid of C. difficile. Of the 60 animals used in this work, it was possible to isolate C. difficile from the feces of 12 (20%), five (41.7%) of the diarrheal group and seven (58.3%) of the group without diarrhea. When interpreting the data from the qualitative detection of toxins A/B of C. difficile by ELISA, it showed that of the 60 animals analyzed, 10 (16.7%) allowed the identification of toxins. Of these 10 piglets, seven (70%) were from the diarrheal group and the other three (30%) from the group without diarrhea. At necropsy, 13 piglets (21.67%) had mesocolon edema, the main macroscopic change associated with infection of C. difficile. Of the 13 animals that had mesocolon edema, eight (61.54%) were from the diarrheal group and the other five (38.46%) without diarrhea. Microscopically, 16 animals showed histopathological lesions characterized by inflammatory lesions. Of these 16 piglets, seven (43.75%) had clinical cases of diarrhea and other nine (56.25%) were non-diarrheal. Of the 60 animals used in this study, it was possible to isolate characteristic colonies of C. perfringens from the feces of 50 (83.3%). With the characterization of these samples, we found that all were C. perfringens type A. Of these 50 strains of C. perfringens type A, 23 (46%) were isolated from animals with diarrhea and 27 (54%) isolated from apparently healthy animals. In 31 (62%) strains of C. perfringens type A isolated, we detected the presence of cpb2 gene, encoding the beta-2 toxin. The result of the sequencing of the cpb2 gene revealed that only 10 samples, eight from the diarrheic group and two from the non-diarrheic group, have a mutation in the gene cpb2 that generated a truncated protein without effective action. The other 21 samples, 12 from the diarrheal group and nine from the group without diarrhea, didnt have the mutation, thereby allowing normal translation of beta-2 toxin. There was not a single sample of C. perfringens type C. The minimal inhibitory concentration (MIC) of C. perfringens type A strains has shown that amoxicillin, ceftiofur and narasin were the antimicrobials which had the best inhibitory activity. The polyvalent vaccine containing recombinant toxoids recombinant alpha, beta and native A/B was effective in stimulating neutralizing antibodies in animal studies.
