dc.description.abstract | Two hemorrhagic factors, mutalysin I and II (LHF-I and LHF-II) were isolated from bushmaster snake (Lachesis muta muta) venom by gel filtration and ion exchange chromatography. The purified proteins represent 12 and 1% respectively of the hemorrhagic activity in the whole venom. The mutalysins were characterized as metalloendopeptidases and contain zinc and calcium. LHF-I (100 kDa, two subunits) and LHF-II (22,4 kDa, single chain) hydrolyze the Aa > Bb of fibrinogen without clot formation. The specific fibrinogenolytic activity was estimated as 25.5 and 13 mg of fibrinogen/min/mg protein for LHF-II and LHF-I respectively. In vitro, the mutalysins showed direct fibrinolytic activity. Analysis by SDS-PAGE of fibrin hydrolysis by both metalloenzymes showedthat LHF-II (0.22 mM) digested completely the a, b and g chains. On the other hand, LHF-I (0.56 mM) hydrolyzes selectively the a chain of fibrin, leaving the b and g chains unaffected. The mutalysins I and II are fibrinogenases which induce hemorrhage without clot formation. In contrast with the plasminogen activator - based thrombolytic agents such as streptokinase, LHF-I and LHF-II do not activate plasminogen. Mutalysin II has no effect on protein C activation and does not inhibit platelet aggregation in human PRP stimulated by ADP or collagen. In addition to hemorrhagic effect by direct proteolysis on capillarywalls, the hemorrhagic toxins can digest other protein components of thehaemostatic system and also activate endogenous proteinases. | pt_BR |
dc.description.resumo | Dois fatores hemorrágicos, mutalisina I e II (LHF-I e LHF-II), foram isolados do veneno de Lachesis muta muta por processos cromatográficos convencionais de filtração molecular e troca iônica, correspondendo a 12 e 1%, respectivamente, da atividade hemorrágica do veneno. Estes componentes foram caracterizados como metaloendoproteinases contendo zinco e cálcio. LHF-I (100 KDa, dímero) e LHF-II (22,5 KDa, cadeia polipeptídica simples) hidrolisam o fibrinogênio com especificidade Aa > Bb, gerando produtos solúveis e fibrinogênio incoagulável pela trombina. A atividade fibrinogenolíticaespecifíca foi estimada em 25,5 e 13,0 mg de fibrinogênio/min/mg de enzima para LHF-II e LHF-I, respectivamente. In vitro, as mutalisinas dissolvem coágulos de fibrina bovina e humana por proteólise direta. A análise por SDSPAGE da hidrólise de fibrina pelas duas metaloproteinases mostra que o fator LHF-II (0,22 mM) digere completamente as cadeias a, b e g. Em contraste, o fator LHF-I (0,56 mM) hidrolisa seletivamente a cadeia a da fibrina sem afetar as cadeias b e g. As mutalisinas I e II pertencem ao grupo de fibrinogenasesque produzem hemorragia sem formação de coágulo. Diferentemente dos agentes usados na terapia trombolítica, tais como a streptokinase, LHF-I e LHF-II não ativam o plasminogênio. A mutalisina II não ativa a proteína C plasmática e não inibe a agregação plaquetária induzida pelo colágeno ou ADP. As metaloproteinases hemorrágicas, além de ação direta sobre as paredes dos capilares devem afetar outros componentes do sistema hemostático, cuja evolução pode levar à hemorragia fatal e aos eventos tromboembólicos observados nos acidentes causados por -serpentes-. | pt_BR |
