Title: Identificação e caracterização de peptídeos bioativos através de Phage display usando genoma completo de Corynebacterium pseudotuberculosis

Abstract: A linfadenite caseosa (LC) é uma doença causada por Corynebacterium pseudotuberculosis, que acomete principalmente caprinos e ovinos. Esta enfermidade é responsável por perdas econômicas significativas relacionadas à produtividade dos animais infectados. E, apesar de sua importância, atualmente não existem vacinas, diagnóstico e tratamentos satisfatórios para LC. No presente trabalho, foi utilizada a ferramenta Phage Display a qual seleciona peptídeos através de bacteriófagos geneticamente modificados para expressar aleatoriamente sequências de peptídeos na sua superfície. Estas bibliotecas têm demonstrado sucesso em numerosas aplicações, como mapeamento e mimetismo de antígenos reconhecidos por anticorpos, desenvolvimento de vacinas, e em testes de imunogenicidade. Sendo assim, este trabalho teve como objetivos selecionar e identificar pela técnica de Phage Display peptídeos ligantes específicos para proteínas secretadas e/ou excretadas e totais de C. pseudotuberculosis linhagem 1002. Para isso foi utilizada uma biblioteca comercial de 12 peptídeos lineares (Ph.D.-12 mer - New England Biolabs - NEB) e após três ciclos de seleção foram obtidos 366 peptídeos, sendo que 116 são seqüências distintas. Foram realizadas análises in silico desses peptídeos com o proteoma predito de C. pseudotuberculosis, onde apresentaram similaridade com proteínas envolvidas na sinalização, nutrição, metabolismo, patogênese bacteriana e fatores de transcrição entre outras. A reatividade e especificidade dos clones isolados foram testadas por ELISA e Spot síntese. Através de imunoensaios para ambas as metodologias foi possível selecionar 12 peptídeos (dos 116 identificados por Phage Display), entre os quais três foram sintetizados, agregados a duas formulações distintas de lipossomas, uma com fosfatidilcolina hidrogenada (HPC) e a outra com fosfatidilcolina de soja (SPC), e utilizados em ensaios de imunização de camundongos BALB/c. A imunização com os peptídeos sintetizados foi capaz de induzir uma elevada resposta imune do tipo Th1 antes da infecção. Após o desafio, foi observada uma produção elevada de IFN- nos animais imunizados tanto com peptídeo/SPC quanto com peptídeo/HPC, e os níveis de IL-10 foram mais elevados nos animais imunizados com peptídeo/SPC. Em contraste, camundongos dos grupos controle exibiram baixos níveis de IFN e de IL-10. A identificação de novos antígenos ou genes específicos de C. pseudotuberculosis pode prover novos biomarcadores específicos potencialmente antigênicos para a obtenção de antígenos candidatos à vacina e diagnósticos subclínicos mais acurados para o controle e erradicação da LC.
Abstract: The Caseous Lymphadenitis (CL) is a disease caused by Corynebacterium pseudotuberculosis, which affects mainly sheep and goats. This illness is responsible for significant economic losses related to productivity of animals. Currently, there is no satisfactory diagnosis for CL. In this work the tool Phage Display was used that selects peptides using bacteriophages to express random sequences of peptides on their surface. Phage Display has been used successfully in many applications such as mapping and mimicry antigens, vaccine development and immunogenicity tests. The main goal of the study was to perform the selection and identification of mimotopes peptides of secreted and /or excreted and total C. pseudotuberculosis 1002 proteins through Phage Display technique. A commercial librarary of 12 linear peptides (Ph.D.-12 mer - New England Biolabs - NEB) was used. After three cycles of selection, 366 peptides were obtained, including 116 distinct sequences. In silico analysis of the peptides obtained in Phage Display were tested against the predicted proteome of C. pseudotuberculosis 1002. The indentified proteins showed similarity to protein involved in signaling, nutrition, metabolism, bacterial pathogenesis and transcription factors. The reactivity and specificity of isolated clones were tested by ELISA and Spot synthesis. Through immunoassays for both methods, was possible to select 12 peptides among them three were synthesized, bound to two different formulations of liposomes, with a hydrogenated phosphatidylcholine (HPC) and soy phosphatidylcholine (SPC), the three peptide were used for testing and immunization of BALB/c. Immunization with the peptides 1 and 2 was able to induce a high Th1 immune response before infection. After the challenge, we observed a high production of IFN- in animals immunized with either peptide/SPC or peptide/HPC, and the levels of IL-10 were higher in animals immunized with peptide/SPC only. In contrast, control groups of mice exhibited low levels of IFN- and IL-10. The identification of new antigens or C. pseudotuberculosis specific genes can potentially provide new specific biomarkers with antigens for obtaining vaccine candidate antigens and sub-clinical diagnosis tests for more accurate control and eradication of the CL.
