Abstract: | A implantação embrionária inclui uma sequência de eventos complexos, com a participação de muitas moléculas mediadoras da interação entre o embrião e o endométrio materno. O presente estudo avaliou a expressão de -inibina e betaglicano na fase secretora do endométrio de mulheres em propedêutica de infertilidade submetidas à fertilização in vitro (FIV), e a relação desses marcadores com a taxa de implantação embrionária. O estudo caso-controle incluiu 52 mulheres submetidas a FIV com transferência embrionária, divididas em dois grupos: casos com falha de implantação (n=33) e controles com gestação clínica confirmada (n=19). A expressão gênica endometrial de -inibina e betaglicano foi avaliada na fase secretora do ciclo menstrual anterior à FIV por PCR em tempo real, através do protocolo do Power SYBR® Green Master Mix. Os resultados indicaram um aumento significativo da expressão gênica de -inibina e betaglicano no endométrio das mulheres com falha de implantação. As áreas sob as curvas ROC para predizer falha de implantação foram, respectivamente, 0,692 e 0,678 para -inibina e betaglicano. Os resultados do presente estudo sugerem que a expressão aumentada de mRNA de -inibina e betaglicano na fase secretora do endométrio pode estar associada à ocorrência de falha de implantação em mulheres submetidas a FIV. |
Abstract: | Embryo implantation involves a complex sequence of events, and a large amount of molecules have been postulated to be involved in the interaction of embryo and endometrium. This study evaluated the endometrial expression of -inhibin and betaglycan in the mid-secretory phase of women scheduled to IVF and tested whether these markers are associated with implantation failure. We conducted a nested case-control study including 52 women submitted to in vitro fertilization (IVF) and embryo transfer, divided into two groups: cases with implantation failure (n=33) and controls with confirmed clinical pregnancy (n=19). Endometrial -inhibin and betaglycan gene expression was evaluated in the mid-secretory phase of the natural menstrual cycle immediately before IVF, using real time PCR. We found a higher gene expression of -inhibin and betaglycan in implantation failure patients compared to confirmed clinical pregnancy patients. The areas under receiver operating characteristics curves for prediction of implantation failure in this context were, respectively, 0.692 and 0.678 for -inhibin and betaglycan. The present results suggest that high expression levels of -inhibin and betaglycan transcripts in secretory phase endometrium may associated to a implantation failure in IVF treatment. |
