Abstract: | Listeria monocytogenes é uma bactéria Gram positiva, intracelular facultativa e invasiva que tem tropismo para a placenta, e causa morbilidade e mortalidade fetal em várias espécies de mamíferos. Embora a infecção por L. monocytogenes e L. ivanovii sejam conhecidas como causa importante de aborto e falhas reprodutivas em bovinos, a patogênese da listeriose materno-fetal nesta espécie é pouco conhecida. Este estudo utilizou o modelo de explante de membrana corioalantóidea bovina para investigar a cinética de infecção por L. monocytogenes, L. ivanovii e L. innocua em células trofoblásticas da espécie bovina por até 8 horas pós-infecção. L. monocytogenes e L. ivanovii foram capazes de invadir e multiplicar em células trofoblásticas sem causar morte celular ou induzir a expressão de genes pró-inflamatórios. Embora L. innocua tenha sido incapaz de se multiplicar em células trofoblásticas da espécie bovina e induzir a transcrição do mediador pró-inflamatório CXCL6. Este estudo demonstrou pela primeira vez a susceptibilidade das células trofoblásticas bovinas para infecção por L. monocytogenes e L. ivanovii. O modelo de explante de membrana corioalantóidea bovina foi adequado para estudar os mecanismos moleculares de patogênese da Listeria spp. em células trofoblásticas bovina, o que pode nos levar a melhor compreensão do aborto induzido por Listeria spp. em bovinos. |
Abstract: | Listeria monocytogenes is a Gram-positive, facultative intracellular and invasive bacterium that has tropism to the placenta, and causes fetal morbidity and mortality in several mammalian species. While infection by L. monocytogenes and L. ivanovii are known as an important cause of abortion and reproductive failure in cattle, the pathogenesis of maternal-fetal listeriosis in this specie is poorly known. This study used the bovine chorioallantoic membrane explant model to investigate the kinetics of L. monocytogenes, L. ivanovii and L. innocua infections in bovine trophoblastic cells for up to 8 hours post infection. L. monocytogenes and L. ivanovii were able to invade and multiply in trophoblastic cells without causing cell death or inducing expression of pro-inflammatory genes. Although L. innocua was unable to multiply in bovine trophoblastic cells, it induced transcription of the proinflammatory mediator CXCL6. This study demonstrated for the first time the susceptibility of bovine trophoblastic cells to L. monocytogenes and L. ivanovii infection. The bovine chorioallantoic membrane explant model proved to be a suitable tool for studying the molecular mechanisms of Listeria spp. pathogenesis in bovine trophoblastic cells, which may lead us to a better understanding of Listeria-induced abortion in cattle. |
