dc.description.abstractBreast cancer is the sixth leading cause of cancer death in the world, and the most frequent type in women. Each year, over 22% of the new cases of cancer in women are breast cancer. Advances in breast cancer detection and treatment have contributed to improve the rate of survival, although mortality rates remain significantly high. Despite all these advances, more efficient diagnostic methods and effective treatments are necessary. The establishment of breast cancer cell lines is an important tool to understand biological processes involved in this disease, as well as the development of potential model of treatment. In the present work, two cell lines, MACL-1 and MGSO-3, from human primary breast cancer were characterized and the effect of paclitaxel on the radiation response of breast cancer cell line was investigated. Characterization of the cell lines included morphological examination that demonstrated a typical epithelial morphology and PCR analyses which showed that both cell lines were telomerase positive. Real time PCR showed that the levels of MUC1 expression in both cell lines were greater than in MDA-MB-231 cell line. Moreover, MACL-1 and MGSO-3 were capable of growing in soft agar culture, which suggests their metastatic potential, and both demonstrated a positive tumorogenic potential in nude mice. These experimental models open new perspectives on the investigation of breast cancer biology and treatment. Our data also showed that pretreatment with paclitaxel enhanced radiation-mediated cell death, promoting caspase-3 activity and apoptosis in breast cancer cell lines. Although, only MACL-1 showed significant difference in clonogenic assays between radiosensivity parameters calculated from irradiated cells, with or without paclitaxel. In conclusion, the results obtained showed that paclitaxel did not induce systematically a radiosensitization and that a synergistic effect was observed in MACL-1 cell line.pt_BR
dc.description.resumoO câncer de mama é o sexto tipo de câncer que mais mata no mundo, sendo o mais freqüente entre as mulheres. A cada ano, cerca de 22% dos novos casos de câncer em mulheres são de mama. Avanços na detecção e tratamento do câncer de mama têm contribuído para melhorar a taxa de sobrevivência, embora a taxa de mortalidade continue alta. Com isso, o desenvolvimento de métodos de diagnósticos mais eficientes e tratamentos mais efetivos continuam sendo necessários. O estabelecimento e caracterização de linhagens de câncer de mama são uma importante ferramenta para o entendimento dos processos biológicos envolvidos nessa doença, assim como para o desenvolvimento de um potencial modelo de tratamento. No presente trabalho, duas linhagens de sítio primário de câncer de mama humano, MACL-1 e MGSO-3, foram caracterizadas e foi também investigado o efeito do paclitaxel na resposta à radiação em linhagens de câncer de mama. A caracterização das linhagens celulares incluiu análise morfológica, que demonstrou uma típica morfologia epitelial e a análise por PCR demonstrou que ambas as linhagens são telomerase positivas. A análise por PCR em tempo real demonstrou que o nível de expressão de MUC1 em ambas as linhagens é maior que aquela da linhagem MDA-MB-231. Além disso, MACL-1 e MGSO-3 apresentaram crescimento independente de ancoragem, que sugere seu potencial metastático, e ambas as linhagens demonstraram ser tumorigênicas em camundongos nude. Estes modelos experimentais abrem novas perpectivas para o estudo da biologia e tratamento do câncer de mama. Nossos dados também demonstraram que o pré-tratamento com paclitaxel aumentou a morte celular, promovendo ativação de caspase-3 e aumentando a apoptose induzida por radiação em linhagens de câncer de mama. Porém, somente MACL-1 demonstrou diferença no ensaio de clonogenicidade entre os parâmetros de radiosensibilização calculados para as células irradiadas, com ou sem paclitaxel. Em conclusão, os resultados obtidos demonstraram que o paclitaxel não induziu uma radiosensibilização sistemática e que um efeito de sinergismo foi observado na linhagem MACL-1.XXpt_BR
