dc.description.abstract | Activin A and follistatin are growth factors produced by several organs and are both measurable in systemic circulation. Human endometrium also expresses activin A and follistatin. The purpose of the present study was to evaluate whether activin A and follistatin are measurable in the menstrual blood, and whether their concentration change in women with dysfunctional uterine bleeding (DUB). Normal cycling women requesting intrauterine contraception (n=15) and a group of women with DUB (n=12)were studied on day 2 of menstrual bleeding. Activin A and follistatin were measured in both menstrual and peripheral sera by using specific ELISA. Activin A concentration in menstrual serum was fourfold higher than in peripheral serum of healthy women (mean ± SE 4.24 ± 0.18 vs. 1.00 ± 0.15 ng/ml) and was significantly lower in women with DUB (2.70 ± 0.42 ng/ml and 0.23 ± 0.02 ng/ml in menstrual and peripheral serum, respectively; p<0.05). Follistatin concentration in menstrual serum was eightfold higher than in peripheral serum of healthy women (3.94 ± 0.49 vs. 0.49 ± 0.04 ng/ml), while was significantly lower in the menstrual serum of women with DUB (1.24 ± 0.22 ng/ml). There was no correlation between concentrations in menstrual serum and peripheral serum. In conclusion, both activin A and follistatin are measurable in high concentration in human menstrual blood and are relatively lower in women with DUB. The quantitative assessment of activin A and follistatin in menstrual serum might be a putative clinical marker of endometrial function. | pt_BR |
dc.description.resumo | A ativina A é um fator de crescimento produzido pelo endométrio e afolistatina é a sua proteína ligante que neutraliza a biodisponibilidade da ativina A. A produção endometrial de ativina A aumenta durante a fase secretora do ciclo menstrual. A proposta do presente estudo foi avaliar se a ativina A e a folistatina são mensuráveis no soro do sangue menstrual, e se essas duas proteínas estão alteradas em mulheres com sangramento uterino disfuncional. Avaliamos 15 mulheres (grupo 1) com ciclos menstruais regulares, que inseriram dispositivo intrauterino (DIU) para contracepção, e comparamos com 12 mulheres com sangramento uterino disfuncional (grupo 2) na unidade Floramar I de atendimento básico da Prefeitura de Belo Horizonte. Ativina A e a folistatina foram medidas no sangue menstrual e periférico usando ensaio-imunoenzimático (ELISA). As concentrações de ativina A foram quatro vezes maiores no soro menstrual que no soro perifériconas mulheres com ciclos normais (média ± desvio padrão 4,24 ± 0,19 vs 1,00 ± 0,15 ng/ml), e foram significantemente menores em mulheres com sangramento uterino disfuncional (2,70 ± 0,42 ng/ml e 0,23 ± 0,03 ng/ml no sangue menstrual e periférico, respectivamente; p<0,001). As concentrações de folistatina foram oito vezes mais altas no soro do sangue menstrual que no periférico das mulheres com ciclos normais (3,94 ± 0,49 vs 0,49 ± 0,05 ng/ml; p<0,001). Os níveis de folistatina foramsignificantemente, mais baixos no soro menstrual das mulheres com sangramento uterino disfuncional (1,24 ± 0,22 ng/ml) comparado com os controles (p<0,01), mas não no soro periférico (0,62 ± 0,09 ng/ml). Não houve nenhuma correlação entre as concentrações dessas proteínas no sangue menstrual e suas respectivas concentrações no soro periférico. Concluindo, ambas ativina A e folistatina são mensuráveis no sangue menstrual, e as concentrações delas são relativamente mais baixas nas mulheres com sangramento uterino disfuncional. Devido o fácil acesso do sangue menstrual que o periférico comparado à biópsia de endométrio, a avaliação quantitativa de ativina A e folistatina no soro menstrual pode ser um importante marcador da função endometrial. | pt_BR |
