Title: | Híbrido de monastrol-H2S : síntese e atividade antiproliferativa contra células de câncer de ovário |

Abstract: | O câncer representa uma das principais causas de morte no mundo. Segundo a OMS, esta doença foi responsável por 8,2 milhões de mortes em 2012 em grande parte causadas por ausência de tratamentos eficazes. Sendo assim, torna-se necessário desenvolver novas substâncias com potencial ação antitumoral. Dentre essas malignidades, o câncer de ovário é caracterizado pela alta mortalidade, pois o diagnóstico geralmente é tardio e o tratamento existente, muitas vezes, não se mostra eficiente. Com isso, na busca por tratamentos alternativos cabe citar dois compostos interessantes, monastrol e ADTOH. O monastrol é um reconhecido inibidor da enzimaEg5 que impede as células de completarem o ciclo de divisão. O ADTOH é uma substância reconhecida por liberar pequenas doses de H25 em meio biológico e apresenta-se como terapia promissora para o tratamento da inflamação. Motivado pelos efeitos peculiares dessas substâncias esse trabalho apresenta a síntese e a avaliação antiproliferativa do híbrido de monastrol-H2S oriundo da união entre oderivado ácido do monastrol e o ADTOH com um rendimento global de 0,4%. Não há na literatura relatos sobre a síntese desse híbrido bem como de suas propriedades antiproliferativos. Os ensaios antiproliferativos foram realizados em linhagem de tumorde ovário humano, TOV-21G (ATCC). Os testes antiproliferativos em células TOV-21G mostraram que o híbrido de monastrol possui propriedades citotóxicas mais parecidas com o monastrol que com oADTOH. Verificou-se ainda que a viabilidade da TOV-21G não é concentração dependente nas concentrações avaliadas (1 pg/mL, 5 pg/mL,10 pg/mL e 15 pg/mL) e que serão necessários outros testes em diferentes concentrações e com um painel tumoral maior para a análise da atividade antitumoral do híbrido. |
Abstract: | Cancer represents one of the mayor causes of death in the world. According to OMS, this illness has been responsible for 8,2 million deaths in 2012, mainly due to the absence of efficient methods and drugs for treatment. Thus, it is necessary to develop new potential antitumor compounds Among these malignancy, the ovarian cancer isresponsible by high mortality, because the diagnosis usually is late and existing treatment, it is often not efficient. In the search for alternative treatment could be mentioned monastrol and ADTOH. Monastrol is a recognized Eg5 enzyme inhibitor that prevents cells to complete the cell division cycle. ADTOH is a compound recognized for releasing H2S in small doses in biological media and shows itself as a promising drugfor inflammation therapy. Motivated by these compounds peculiar effects this study esis and evaluation of hybrid of monastrol-H2S produced from the coupling between the acid monastrol derivative and the ADTOH with a global yield of 0,4%. There is not any report of the synthesis as well about antiproliferative properties of this hybrid. The antiproliferative trials were accomplished in human ovarian tumor lines TOV-21G (ATCC).The antiproliferative test in TOV-21G cells showed that monastrol hybrid had cytotoxic properties more similarly to monastrol than to ADTOH. It was confirmed that the TOV-21G viability is not dose dependent in the evaluated concentrations (1 pg/mL, 5 pg/mL, 10 pg/mL and 15 pg/mL). In addition, it will be necessary another trials with different concentrations and a bigger tumor panel for antitumor activity's hybrid analysis. |
