Abstract: | Bases de Schiff, respectivas aminas secundárias bem como N-metilaminas análogas ao antifúngico butenafina foram sintetizadas. A síntese foi eficiente, simples e rápida para os dois primeiros grupos de compostos mencionados, que foram obtidos com rendimentos globais superiores a 94% em sua maioria. Em contraste, os rendimentos globais para a síntese das N-metilaminas terciárias foram em geral superiores a 66% para as aminas benzílicas, sendo estas melhores substratos que as fenílicas. As bases de Schiff, aminas secundárias e terciárias e respectivos cloridratos foram, então, avaliados quanto ao potencial antifúngico de interesse clínico. Os fungos Cryptococcus gattii ATCC 32608, Aspergillus fumigatus ATCC 16913, Trichophyton interdigitale e Microsporum gypseum foram então estudados para fins de determinação da concentração mínima inibitória (CIM) dos compostos-teste necessária para inibir 100% do crescimento microbiano. As N-metilaminas mostraram-se mais ativas do que as aminas secundárias correspondentes. Os cloridratos, a exemplo de 5b.HCl, 8b.HCl, 2c.HCl e 8c.HCl, e as aminas livres 5b, 5c e 8c foram na maioria dos casos tão ativos quanto o antifúngico-referência (butenafina; CIM < 4 µg.L-1) para três dos quatro fungos testados. Notavelmente, a presença dos grupos naftila e 4-terc-butilbenzila, bem como um menor caráter básico do átomo do nitrogênio em meio aquoso, contribuíram para o aumento da atividade antifúngica dos compostos testados. Os resultados obtidos indicam que os cloridratos 5b.HCl, 8b.HCl, 2c.HCl e 8c.HCl, além de ser sintetizados em rendimentos globais superiores a 67%, mostraram-se promissores antifúngicos contra os fungos C. gattii ATCC 32608, T. interdigitale e M. gypseum, podendo serem utilizados como plataforma para o desenvolvimento de antifúngicos ainda mais potentes. |
Abstract: | Schiff bases, secondary amines and their N-methylamines analogous to antifungal butenafine were synthesized. The synthesis was efficient, simple and rapid for the first two groups of compounds mentioned, which were obtained with global yields higher than 94% at most. In contrast, overall yields for the synthesis of N-tertiary methylamines weregenerally above 66% for the benzylic amine, which are better substrates than the phenylic amines. Schiff bases, secondary and tertiary amines and their hydrochlorides were then evaluated for antifungal potential clinical interest. The fungus Cryptococcus gattii ATCC 32608, ATCC 16913 Aspergillus fumigatus, Trichophyton interdigitale and Microsporumgypseum were then studied in order to determine the minimum inhibitory concentration (MIC) of test-compounds required to inhibit 100% of microbial growth. The N-methylamines were more active than the corresponding secondary amines. The hydrochlorides, by way of example5b.HCl, 8b.HCl, 2c.HCl, 5c.HCl and 8c.HCl, and the free amines 5b, 5c and 8c were in most cases as active as antifungal reference (butenafine; MIC < 4 ug.L-1) for three of the four fungi tested. Notably, the presence of the naphthyl and 4-tert-butylbenzyl group, as well as lower basic character of the nitrogen atom in aqueous medium, contributed to increasing theantifungal activity of the tested compounds. The results indicate that the hydrochloride 5b.HCl, 8b.HCl, 2c.HCl, 5c.HCl and 8c.HCl, further it is synthesized in overall yields greater than 67%, have shown promising antifungal against the fungus C. gattii ATCC 32608, T. interdigitale and M. gypseum, may be used as a platform for the development of even morepotent antifungals. |
