Resumen: | Os objetivos deste estudo foram avaliar o efeito do concentrado autologo de plaquetas (CAP) sobre a osteoartrose (OA) induzida em coelhos, por meio de exames clinicos, radiograficos, ultrassonograficos, laboratoriais e artroscopicos, e determinar a concentracao dos fatores de crescimento derivado das plaquetas (PDGF) e transformador (TGF-) no plasma de coelhos tratados com CAP e com Ringer Lactato. Foram empregados 14 coelhos da raca Nova Zelandia com massa corporal media de 3,0 kg submetidos a ruptura do ligamento cruzado cranial (LCCr) e posterior substituicao por auto-enxerto de fascia lata, guiados por videoartroscopia. Os animais foram distribuidos em dois grupos de sete e receberam injecoes intra-articulares de CAP ou de Ringer Lactato. Na avaliacao ortopedica os coelhos tratados com CAP mostraram resultados (escores) significativamente superiores (p<0,05) ao grupo controle quanto a sensibilidade dolorosa, presenca de efusao articular e apoio do membro no momento M3. Houve diferenca estatisticamente significativa (p<0,05) nos angulos de flexao e extensao do membro posterior direito (MPD) entre os grupos no momento M3, com melhores resultados nos animais do grupo CAP. Nao houve diferenca entre os grupos nas avaliacoes radiograficas eultrassonograficas nos tempos avaliados. Foi observado entre os grupos, diferenca estatisticamente significativa (p<0,05) na avaliacao do liquido sinovial somente em relacao ao volume e a proteina no M3. Verificou-se na contagem celular, diferenca estatisticamente significativa (p<0,05) entre o CAP e o sangue total nos momentos M2 e M3. O volume plaquetario medio (MPV) entretanto, foi semelhante. A concentracao do PDGF foi estatisticamente maior (p<0,05) nos coelhos tratados com CAP no M3 em relacao ao grupo controle, mas nao houve diferenca estatistica na concentracao do TGF-1 entre os grupos em nenhum dos momentos avaliados. Os resultados obtidos permitem concluir que o metodo empregado para concentrar plaquetas inativas de coelhos foi efetivo e que o CAP tem efeito favoravel em articulacoes acometidas pela osteoartrose (OA). A dosagem dos fatores de crescimento PDGF e TGF-1, pode ser usada como marcadores biologicos para avaliar o processo degenerativo. |
Abstract: | The objectives of this study was to evaluate the effect of the autologous platelet concentrate (APC) on osteoarthritis (OA) induced in rabbits, through clinical, radiographic, ultrasonographic, laboratorial and arthroscopic evaluations, and to determinate the platelet derived growth factor (PDGF) and of transforming growth factor 1 (TGF-1) concentrations in plasma of rabbits treated with APC or Ringer Lactato. Were used 14 New Zealand rabbits with average body weight of 3,0 kg submitted to cranial cruciate ligament (LCCr) resection and substitution with fascia lata auto-graft guided by arthroscopy. The rabbits were distributed in two groups of seven animals each and received intra-articular injections of APC or Ringer Lactato. In the orthopedic evaluation the APC group showed significantly better results (p<0,05) than the control group rabbits in relation to the pain sensitivity, presence of articular effusion and member support and position in moment M3. It was observed significant statistically differences (p<0,05) in the flexion and extension angles in the right hindlimb between groups in the M3. It was not observed significant differences between the groups in the radiographic and ultrassonographic evaluation. It was observed between the groups significant statistically difference (p<0,05) in the synovial fluid evaluation just in relation to volume and protein parameters in the moment M3. It was verificated in the cellular counts, significant statistically diference (p<0,05) between the APC and the total blood in the moments M2 and M3. The medium plaquetary volume (MPV), however, was simillar. The PDGF concentration was statistically higer (p<0,05) in ACP group in the M3 in relation to the control group, but there was not statistically significant difference in the TGF-1 concentration between groups on any of the avaliated moments. The obtained result allow to conclude that the employed method to concentrate inactive platelets of rabbits was effective and that ACP have a favorable effect in joints affected by osteoarthritis (OA). The dosage of growth factors PDGF e TGF-1, can be used as a biological marker to evaluate the degenerative process |
