| Abstract: | Muitos estudos têm sido realizados em todo o mundo a respeito da resposta imunológica de cavalos (Equus caballus) à Anemia Infecciosa Eqüina (AIE), no entanto existe pouca informação a respeito da resposta imunológica de muares (Equus caballus X Equus asinus) e asininos (Equus asinus) naturalmente infectados pelo VAIE. O objetivo desse trabalho foi comparar o perfil sorológico e a detecção do vírus da AIE em asininos, muares e eqüinos naturalmente infectados e avaliar a importância destas espécies como mantenedoras da AIE nos rebanhos brasileiros. No presente estudo, foi coletado sangue para obtenção de soro e células mononucleares periféricas (PBMC) de 124 cavalos, 119 muares e 75 asininos, perfazendo um total de 318 animais. Foi extraído DNA do PBMC de 114 cavalos, 79 muares e 35 asininos. Todas as amostras foram testadas através da Imunodifusão em Gel de Agar (IDGA) e Ensaio imunoenzimatico (ELISA). O DNA foi submetido à reação nested da polimerase em cadeia (nPCR). Nos eqüinos ocorreu a melhor correlação entre IDGA e ELISA do que nos muares. O mesmo foi observado entre a IDGA e a nPCR. Quando se comparou a correlação entre ELISA e a nPCR, o melhor resultado foi observado para amostras de eqüinos novamente. O número de amostras de asininos foi insuficiente para se fazer uma análise estatística, mas os resultados mostraram que dez asininos, negativos na IDGA, foram positivos no ELISA e sete positivas na nPCR. As duas amostras positivas na IDGA foram também positivas no ELISA e na nPCR. De acordo com os resultados obtidos, pode-se concluir que as respostas sorológicas e viremia dos muares são diferentes das respostas dos eqüinos. A concordância entre a IDGA, ELISA e PCR em amostras de eqüinos foi superior aos resultados obtidos em amostras de muares. |
| Abstract: | A lot of studies about horses (Equus caballus) immune responses to equine infectious anemia virus (EIAV) have been done all over the world, but there are little information concerning immune responses of mule (Equus caballus X Equus asinus) and donkey (Equus asinus) to natural infection with EIAV. The aim of this work was to compare the serological and virus detection in asinus, mules and equines naturally infected with EIAV, and evaluate the importance of mules and asinus to maintain the EIAV in the Brazilian herds. In the present study sera and peripheral blood mononuclear cells (PBMC) samples were collected of 124 horses, 119 mules and 75 asinus, in a total of 318 samples. DNA of PBMC was extracted from 114 horses, 79 mules and 35 asinus. All samples were tested by Agar Gel Immunodiffusion (AGID) and Enzyme-linked Immunosorbent Assay (ELISA). DNA was submitted to nested polymerase chain Reaction (nPCR). The correlation to AGID and ELISA for equines was better than mules. The same was observed between AGID and nPCR. When the correlation between ELISA and nPCR was compared, the best correlation was observed in equines again. The number of asinus was not significant to statistic evaluation but results showed that 10 negative asinus in AGID were positive in ELISA and 7 were positive in nPCR. Two positive asinus in AGID were positive in ELISA and nPCR. These results showed that serological responses and virus detection in mules might be different from the responses in equines. The concordance between AGID, ELISA and PCR tests were better in equine than in mules. |
