Abstract: | Purpose Studies conducted since 1969 have shown that the release of serotonin (5-HT) in the dorsal horn of the spinal cord contributes to opioid analgesia. In the present study, the participation of the opioidergic system in antinociceptive effect serotonin at the peripheral level was examined. Methods The paw pressure test was used with mice (Swiss, males from 35 g) which had increased pain sensitivity by intraplantar injection of PGE2 (2 μg). Serotonin (250 ng), administered locally to the right paw of animals, produces antinociception in this model. Results The selective antagonists for mu, delta and kappa opioid receptors, clocinnamox clocinnamox (40 μg), naltrindole (60 μg) and nor-binaltorfimina (200 μg), respectively, inhibited the antinociceptive effect induced by serotonin. Additionally, bestatin (400 μg), an inhibitor of enkephalinases that degrade peptides opioids, enhanced the antinociceptive effect induced by serotonin (low dose of 62.5 ng). Conclusions These results suggest that serotonin possibly induce peripheral antinociception through the release of endogenous opioid peptides, possible from immune cells or keratinocytes. |
Abstract: | Objetivo Estudos realizados desde 1969 mostraram que a liberação de serotonina (5-HT) no corno dorsal da medula espinhal contribui para a analgesia opióide. No presente estudo, foi examinada a participação do sistema opioidérgico no efeito antinociceptivo da serotonina em nível periférico. Métodos O teste de pressão da pata foi utilizado em camundongos (suíços, machos a partir de 35 g) que apresentaram aumento da sensibilidade à dor pela injeção intraplantar de PGE2 (2 μg). A serotonina (250 ng), administrada localmente na pata direita dos animais, produz antinocicepção neste modelo. Resultados Os antagonistas seletivos dos receptores opióides mu, delta e kappa, clocinamox clocinamox (40 μg), naltrindol (60 μg) e nor-binaltorfimina (200 μg), respectivamente, inibiram o efeito antinociceptivo induzido pela serotonina. Além disso, a bestatina (400 μg), um inibidor de encefalinases que degradam peptídios opióides, potencializou o efeito antinociceptivo induzido pela serotonina (dose baixa de 62,5 ng). Conclusões Esses resultados sugerem que a serotonina possivelmente induz a antinocicepção periférica através da liberação de peptídeos opióides endógenos, possivelmente de células imunes ou queratinócitos. |
