Title: | Influence of Long-Term Salt Diets on Cardiac Ca2+ Handling and Contractility Proteins in Hypertensive Rats |

Abstract: | BACKGROUND High sodium intake contributes to the pathogenesis of hypertension and adversely affects cardiac function. Conversely, sodium reduction is associated with a blood pressure decrease and improved cardiovascular function. However, the mechanisms that underlie the cardiac effects induced by salt intake in hypertension have not been fully elucidated. Ca2+ handling is critical for efficient myocardial function; thus, we aimed to investigate the long-term effects of diets with different salt contents on cardiac function and Ca2+ handling proteins in spontaneously hypertensive rats (SHRs). METHODS Cardiac function was evaluated by catheterization. Ca2+ handling and contractile proteins were evaluated by immunoblotting in hearts from SHRs fed for 6 months with diets containing high (HS, 3%), low (LS, 0.03%), or normal salt content (NS, 0.3%). Diets were introduced immediately after weaning. Tail cuff pletismography was assessed at the 3rd and 7th months of follow-up. RESULTS Compared to the NS group, the HS group exhibited worsened hypertension, increased cardiac expression of β-myosin heavy chain (MHC), a decreased α/β-MHC ratio and reduced expression of both phospholamban (PLB) and Na+/Ca2+ exchanger (NCX). LS intake attenuated the blood pressure increase and left ventricle hypertrophy, slightly decreased the cardiac contractility and relaxation index, and increased the α/β-MHC ratio. These effects were accompanied by increased cardiac PLB expression and decreased Ca2+ L-type channel and NCX expression. CONCLUSIONS These findings indicate that the modulation of Ca2+ handling may be one of the molecular mechanisms underlying the effect of salt intake on myocardial function in hypertension. |
Abstract: | FUNDO A alta ingestão de sódio contribui para a patogênese da hipertensão e afeta negativamente a função cardíaca. Por outro lado, a redução de sódio está associada a uma diminuição da pressão arterial e melhora da função cardiovascular. No entanto, os mecanismos subjacentes aos efeitos cardíacos induzidos pela ingestão de sal na hipertensão não foram totalmente elucidados. O manuseio do Ca2+ é crítico para a função miocárdica eficiente; assim, nosso objetivo foi investigar os efeitos a longo prazo de dietas com diferentes teores de sal sobre a função cardíaca e proteínas de manipulação de Ca2+ em ratos espontaneamente hipertensos (SHRs). MÉTODOS A função cardíaca foi avaliada por cateterismo. A manipulação de Ca2+ e proteínas contráteis foram avaliadas por immunoblotting em corações de SHRs alimentados por 6 meses com dietas contendo alto (HS, 3%), baixo (LS, 0,03%) ou teor de sal normal (NS, 0,3%). As dietas foram introduzidas imediatamente após o desmame. A pletismografia do manguito da cauda foi avaliada no 3º e 7º meses de seguimento. RESULTADOS Comparado ao grupo NS, o grupo HS apresentou piora da hipertensão, aumento da expressão cardíaca da cadeia pesada da β-miosina (MHC), diminuição da razão α/β-MHC e redução da expressão tanto do fosfolambam (PLB) quanto do trocador Na+/Ca2+ (NCX ). A ingestão de LS atenuou o aumento da pressão arterial e a hipertrofia do ventrículo esquerdo, diminuiu levemente a contratilidade cardíaca e o índice de relaxamento e aumentou a relação α/β-MHC. Esses efeitos foram acompanhados por aumento da expressão de PLB cardíaco e diminuição do canal do tipo L de Ca2+ e expressão de NCX. CONCLUSÕES Esses achados indicam que a modulação da manipulação de Ca2+ pode ser um dos mecanismos moleculares subjacentes ao efeito da ingestão de sal na função miocárdica na hipertensão. |
