Abstract: | A gota é uma doença inflamatória crônica causada pela deposição de cristais de monourato de sódio (MSU) nas articulações, na qual, desencadeia uma resposta inflamatória caraterizada pelo influxo de neutrófilos, ativação do complexo intracelular de proteínas do inflamassoma e hiperalgesia articular intensa. A prevalência da gota tem aumentado mundialmente nos últimos anos, e, ainda não possui tratamento eficaz. A terapia anti- inflamatória pode ser uma estratégia alternativa, pois detém o início da resposta inflamatória aguda ao MSU, entretanto ela não resolve o problema em si. Considerando as terapias anti- inflamatórias e estratégias alternativas, o peptídeo P140 é capaz de atuar (modulando células do sistema imunológico e a autofagia celular), apresentando efeitos benéficos para o tratamento do Lúpus, também uma doença inflamatória crônica. Assim, o presente trabalho tem como objetivo principal avaliar os efeitos do tratamento com o peptídeo P140 no modelo experimental murino de gota. Pode-se observar que ambas estratégias (pré e pós-tratamento) com o peptídeo sintético P140 reduziram o recrutamento de neutrófilos para a cavidade articular, a hipernocicepção (dor) e a lesão tecidual nos joelhos dos camundongos tratados e estimulados com injeção intra-articular de MSU. Para investigar melhor os mecanismos envolvidos, avaliamos in vitro, a produção de espécies reativas de oxigênio (ROS) em macrófagos estimulados com MSU. Macrófagos derivados de medula óssea (BMDM) estimulados com o peptídeo P140 apresentaram menor produção de ROS, quando comparados com aqueles que não receberam o tratamento, sugerindo menor ativação do complexo de proteína inflamassoma. Em todos os experimentos utilizamos também como controle do peptídeo P140 o peptídeo truncado sC140, afim de melhor elucidar os mecanismos de ação do P140. Sendo assim, concluímos que o tratamento com o peptídeo P140 apresentou efeitos imunomoduladores importante no modelo de gota experimental, sugerindo ser uma estratégia terapêutica interessante para o tratamento desta patologia. |
Abstract: | Gout is a chronic inflammatory disease caused by the deposition of monosodium urate (MSU) crystals in joints, triggering an inflammatory response mark by increasing neutrophils in joints, inflammasome-dependent autophagy activation, and increase of hyperalgesia. The gout prevalence has grown, and this pathology does not have any effective treatment. Anti- inflammatory therapy is the most used strategy because it can stop at the beginning of acute MSU inflammatory response; however, it does not solve the problem. The P140 peptide can modulate the immune system and the autophagy response, presenting beneficial effects in lupus treatment, also a chronic inflammatory disease. Thus, the present work aims to elucidate the P140 treatment effects in a mice model of gout. Both therapeutic strategies, pre and post- treatment, decreased the neutrophil recruitment in joints, the hipernocyception, and the tissue injury in the treated mice. To better investigate the pathways and mechanisms involved, we evaluate in vitro the species reactive oxygen (ROS) production from macrophages stimulated with MSU. The P140 treatment group shows decreased ROS production than the no-treatment group, suggesting a minor inflammasome activation in these cells. In all experiments, we use a control peptide named sC140 to clarify the P140 mechanisms. Therefore, we conclude the P140 treatment show immunomodulatory effects in this mice model of gout, and this suggests this peptide be an attractive therapeutic strategy to treats this pathology. |
