Title: | Cyclic AMP regulates key features of macrophages via PKA: recruitment, reprogramming and efferocytosis |

Abstract: | Macrophages are central to inflammation resolution, an active process aimed at restoring tissue homeostasis following an inflammatory response. Here, the effects of db-cAMP on macrophage phenotype and function were investigated. Injection of db-cAMP into the pleural cavity of mice induced monocytes recruitment in a manner dependent on PKA and CCR2/CCL2 pathways. Furthermore, db-cAMP promoted reprogramming of bone-marrow-derived macrophages to a M2 phenotype as seen by increased Arg-1/CD206/Ym-1 expression and IL-10 levels (M2 markers). Db-cAMP also showed a synergistic effect with IL-4 in inducing STAT-3 phosphorylation and Arg-1 expression. Importantly, db-cAMP prevented IFN-γ/LPS-induced macrophage polarization to M1-like as shown by increased Arg-1 associated to lower levels of M1 cytokines (TNF-α/IL-6) and p-STAT1. In vivo, db-cAMP reduced the number of M1 macrophages induced by LPS injection without changes in M2 and Mres numbers. Moreover, db-cAMP enhanced efferocytosis of apoptotic neutrophils in a PKA-dependent manner and increased the expression of Annexin A1 and CD36, two molecules associated with efferocytosis. Finally, inhibition of endogenous PKA during LPS-induced pleurisy impaired the physiological resolution of inflammation. Taken together, the results suggest that cAMP is involved in the major functions of macrophages, such as nonphlogistic recruitment, reprogramming and efferocytosis, all key processes for inflammation resolution. |
Abstract: | Os macrófagos são fundamentais para a resolução da inflamação, um processo ativo que visa restaurar a homeostase tecidual após uma resposta inflamatória. Aqui, os efeitos do db-cAMP no fenótipo e função dos macrófagos foram investigados. A injeção de db-cAMP na cavidade pleural de camundongos induziu o recrutamento de monócitos de maneira dependente das vias PKA e CCR2/CCL2. Além disso, o db-cAMP promoveu a reprogramação de macrófagos derivados da medula óssea para um fenótipo M2, conforme observado pelo aumento da expressão de Arg-1/CD206/Ym-1 e níveis de IL-10 (marcadores M2). Db-cAMP também mostrou um efeito sinérgico com IL-4 na indução da fosforilação de STAT-3 e expressão de Arg-1. É importante ressaltar que db-cAMP impediu a polarização de macrófagos induzida por IFN-γ/LPS para tipo M1, conforme mostrado pelo aumento de Arg-1 associado a níveis mais baixos de citocinas M1 (TNF-α/IL-6) e p-STAT1. In vivo, db-cAMP reduziu o número de macrófagos M1 induzidos pela injeção de LPS sem alterar os números de M2 e Mres. Além disso, db-cAMP aumentou a eferocitose de neutrófilos apoptóticos de maneira dependente de PKA e aumentou a expressão de anexina A1 e CD36, duas moléculas associadas à eferocitose. Finalmente, a inibição da PKA endógena durante a pleurisia induzida por LPS prejudicou a resolução fisiológica da inflamação. Tomados em conjunto, os resultados sugerem que o cAMP está envolvido nas principais funções dos macrófagos, como recrutamento não flogístico, reprogramação e eferocitose, processos-chave para a resolução da inflamação. |
