Resumen: | A atividade antimicrobiana in vitro de extratos e de compostos fenólicos e nitrogenados do café (ácido caféico, ácido clorogênico, ácido protocatéico, cafeína e trigonelina) isolados ou em associação sobre 15 espécies de bactérias, duas de leveduras e uma de fungo filamentoso foi investigada pelo método de difusão em ágar. Observou-se atividade antibacteriana, com destaque para Salmonella enterica sorovar Typhimurium, Escherichia coli, Staphylococcus aureus e Streptococcus mutans. Não foi verificada atividade antifúngica dos extratos e compostos estudados. Foi observado efeito positivo da associação do extrato de café com a cafeína sobre Bacillus subtilis. Não houve influência significativa de diferentes marcas de café, do tipo de solvente extrator e do modo de preparo na inibição de S. mutans. Estudo cinético utilizando o método turbidimétrico indicou significativa atividade sobre S. mutans em níveis de cafeína 4,0 mg/mL. As concentrações inibitórias de 50% e 90% das substâncias sobre S. enterica, calculadas pelo método de diluição em placa de microtítulo, indicaram maiores atividades antimicrobianas do ácido protocatéico, cafeína e trigonelina. Estudo cinético empregando o método turbidimétrico indicou que o ácido protocatéico e a cafeína apresentaram os maiores efeitos antimicrobianos sobre S. enterica em 24 h. O pH interferiu na atividade antimicrobiana das substâncias sobre S. enterica. Elevada atividade antimicrobiana foi obtida em pH 4,5 para cafeína e ácido protocatéico (96%), seguida da trigonelina e ácido clorogênico (80%). Estudos em modelo alimentar constituído de leite desnatado, pH 4,5 e 5,5, 25 ºC/28 dias, inoculado com 106 UFC/mL de S. enterica indicaram maior eficiência conservante do ácido protocatéico, pH 4,5, comparado à cafeína, com inibição de até três ciclos logarítmicos, em relação ao controle. Alterações morfológicas de S. enterica após 24 h de contato com ácido protocatéico e com cafeína foram observadas em microscopias óptica e de força atômica. Pode-se inferir que a cafeína tenha a sua ação antimicrobiana relacionada à divisão celular, com uma possível interferência na formação do septo. |
Abstract: | ANTIMICROBIAL ACTIVITY OF COFFE EXTRACTS, PHENOLICS AND NITROGENOUS COMPOUNDS: IN VITRO EVALLUATION IN A FOOD MODEL. The in vitro antimicrobial activity of coffee extracts and coffee chemical compounds (caffeic acid, chlorogenic acid, protocatechuic acid, caffeine and trigonelline) isolated or in association against 15 bacteria, 2 yeasts and 1 filamentous fungus were investigated by the agar diffusion method. There was antibacterial activity, mainly against Salmonella enterica serovar Typhimurium, Escherichia coli, Staphylococcus aureus and Streptococcus mutans. There was no antifungal activity of the extracts and chemical compounds. The association of caffeine with the coffee extract provided enhanced antimicrobial effect against Bacillus subtilis. The inhibition of S. mutans was not affected by coffee brand, type of solvent and way of preparation. Kinetic studies using the turbidimetric method indicated that significant effect against S. mutans was observed only at 4.0 mg of caffeine/mL. The 50 and 90% inhibitory concentrations using the microtiter plate dilution method and S. enterica indicated that protocatechuic acid, caffeine and trigonelline promoted the highest inhibitory effects. Kinetic studies using the turbidimetric method indicated that protocatechuic acid and caffeine had the higher antimicrobial effects against S. enterica for 24 h. pH affected the antimicrobial activity of the compounds against S. enterica. High inhibition was observed at pH 4.5 for caffeine and protocatechuic acid (96%), followed by trigonelline and chlorogenic acid (80%). The preservative effect of protocatechuic acid and caffeine in a non-fat dry milk food model, pH 4.5 and 5.5, with 106 UFC/mL of S. enterica was investigated for 28 days at 25 ºC. Protocatechuic acid, pH 4.5, showed a higher preservative effect with a three log cycle inhibition compared to the control. However, the antimicrobial activity was reduced by the presence of milk components. Morphological alterations of S. enterica after 24 h contact with protocatechuic acid and caffeine were observed by optic and atomic force microscopy. It can be inferred that the inhibitory effect of caffeine is related to cell division, with a possible interference in the formation of septum. |
