Abstract: | A campilobacteriose genital bovina e uma enfermidade de carater venereo, que leva a infertilidade temporaria e abortos esporadicos em bovinos. Por isso, tem grande importancia em paises como o Brasil, onde ha grandes rebanhos que utilizam a monta natural. Com o objetivo de verificar se a passagem seriada em novilhas ativa proteinas que estao ligadas a patogenia da infeccao e identificar proteinas diferencialmente expressas em amostras de Campylobacter fetus subsp venerealis, foi feito o teste de aderencia e inducao de transcricao de interleucina-8 (IL-8) em celulas epiteliais infectadas bem como a identificacao de proteinas por espectrometria de massa. Verificou-se que a passagem seriada de C. fetus subsp. venerealis em novilhas leva ao aumento gradual de aderencia e induz a transcricao de IL-8 em celulas HeLa. Passagens seriadas do microorganismo in vivo levam ao aumento na expressao de proteinas como: Purine Binding Chemotaxis (CheW), Flagelar Hook associated, Possible Lipoprotein e Methil accepting chemotaxisis, ligadas a aderencia bacteriana, quimiotaxia bacteriana e motilidade importantes na colonizacao e no estabelecimento da infecção. |
Abstract: | The bovine genital campylobacteriosis is a venereal disease, which leads to temporary infertility and sporadic abortion in cattle. So it has great importance in countries like Brazil, where large herds using natural service. In order to verify that the serial passage in heifers active proteins that are linked to the pathogenesis of infection and identify differentially expressed proteins in samples of Campylobacter fetus subsp venerealis, has been tested for adhesion and induction of transcription of interleukin-8 (IL-8) in infected epithelial cells and identification of proteins by mass spectrometry. It was found that serial passage of C. fetus subsp. venerealis heifers gradually leads to increased adhesion and induces transcription of IL-8 in HeLa cells. Serial passages of the microorganism "in vivo" lead to increased expression of proteins such as Chemotaxis Purine Binding (Chew), Hook Associated scourge, Lipoprotein and Possible Accepting chemotaxisis methyl, linked to the bacterial adherence, chemotaxis and motility important in bacterial colonization and the establishment infection. |
