Abstract: | O objetivo deste estudo foi avaliar o efeito da esterilização em autoclave e do armazenamento em glicerol a 85% de osso cortical de cães infectados por Leishmania sp., e avaliar suas características mecânicas e morfométricas. Foram utilizados 43 cadáveres de cães com leishmaniose e 11 cadáveres de cães isentos da doença. Dos animais infectados, foram colhidas 43 diáfises de fêmur direito e 43 amostras da medula óssea correspondente. A diáfise foi dividida em duas, uma parte fragmentada em pequenas partículas e a outra seccionada em dois fragmentos de cerca de 1cm de comprimento para testes mecânicos e estudo morfométrico. A parte fragmentada foi submetida à cultura do parasito antes e após a esterilização em autoclave e armazenamento em glicerol. Amostras da diáfise dos cães negativos foram também submetidos às avaliações mecânica e morfométrica. Ambos os tratamentos foram eficazes para impedir o crescimento do parasito in vitro. Os testes mecânicos não mostraram diferença estatística na tensão de deformação nem no módulo de elasticidade à compressão mecânica entre os grupos de cães infectados e não infectados. Verificou-se na avaliação morfométrica aumento significativo da percentagem de osteócitos na cortical de animais infectados. Concluiu-se que os tratamentos são eficazes na eliminação do parasito e não há alteração de resistência mecânica do osso devido à infecção, sendo viável seu emprego em enxertia. São necessários mais estudos sobre os mecanismos envolvidos no aumento de osteócitos no tecido cortical. |
Abstract: | The aim of this study was to evaluate the effect of autoclaving and storage in 85% glycerol in cortical bone of dogs infected by Leishmania sp., and evaluate their morphological and mechanical characteristics. We used 43 cadavers of dogs with leishmaniasis and 11 cadavers of dogs free from the disease. From the infected animals were harvested 43 diaphysis of the right femur and 43 corresponding bone marrow samples. The shaft was divided in two, one part fragmented into small particles and the other split into two fragments of about 1cm in length for mechanical testing and morphometric study. The fragmentary was subjected to culture the parasite before and after autoclaving and storage in glycerol. Samples of the diaphysis of negative dogs were also subjected to morphometric and mechanical ratings. Both treatments were effective in preventing the growth of the parasite in vitro. The mechanical tests showed no statistical difference in deformation tension or the elastic modulus by mechanical compression between the infected and uninfected groups. There was a significant increase in the morphometric analysis of the percentage of cortical osteocytes in infected animals. It was concluded that the treatments are effective in eliminating the parasite and there is no change in strength of the bone due to infection, therefore beeing viable their use in grafting. More studies are needed on the mechanisms involved in the increase of osteocytes in cortical tissue |
